Von Kossa staining is a histochemical technique used to detect the presence of calcium deposits in tissue samples. It involves the treatment of the sample with silver nitrate, which reacts with the calcium to form silver phosphate or silver carbonate. The stained sample is then exposed to light, causing the silver compounds to be reduced to metallic silver, which appears as a dark brown or black stain. This staining procedure is commonly used in the analysis of mineralized tissues, such as bone and cartilage.
