Xenograft Model of FL-SJC Tumor in SCID Mice

FL‐SJC cells (10 × 106) were inoculated into the left armpits of 4‐week‐old female SCID mice (Gempharmatech Co., Ltd) by subcutaneous injection (n = 3). Animals were marked and checked every two weeks for lump formation. After 6 to 8 weeks, solid tumour mass under the armpits from all of the 3 mice inoculated with FL‐SJC cells was detected. The mice were sacrificed under euthanasia, and tumour masses were resected for biopsy. All animal experiments have been reviewed and approved by the Institutional Animal Care and Use Committee of Jiangsu University.

Male C57BL/6J mice (10- to 12-week-old, 23–25 g) and BALB/c mice were obtained from Beijing Huafukang Bioscience Co. Inc. Il10 deficient (Il10-null) mice (B6/JGpt-I-l10em1Cd4885/Gpt), whole-body Cyp8b1 knockout (Cyp8b1-null) mice and SCID mice were purchased from Gempharmatech. Cre-dependent Cas9 knock in (Cas9LSL+/+) mice were purchased from Jackson Laboratory. Lgr5-EGFP-IRES-creERT2 mice were purchased from Shanghai Model Organisms Center. Pparafl/fl and PparaΔIE mice were described previously (Luo et al., 2019 (link)). Fxr-null, intestinal Fxr-null (FxrΔIE) and hepatic Fxr-null (FxrΔHep) mice were described previously (Kim et al., 2007 (link); Sinal et al., 2000 (link)). Tgr5-null mice were kindly provided by Prof. Xin Xie (Shanghai Institute of Materia Medica). Mice were maintained under a standard 12-h light/12-h dark cycle with water and food provided ad libitum. All experiments were performed according to the institutional ethical guidelines on animal care and approved by the Institute Animal Care and Use Committees at Shanghai Institute of Materia Medica and the National Cancer Institute.
