DSPG, and soybean phosphatidylcholine S75 (S75) were obtained from Lipoid GmbH (USA). N (carbonyl-methoxy PEG 2000 )-1,2-distearoyl-sn-glycero-3-phosphoethanolamine (mPEG 2000 -DSPE) was obtained from Genzyme Corporation (USA). Synthesis and Characterization of Fluorescent Inositol Polyphosphates

All lipids were purchased form Avanti Polar Lipids, Inc. (Alabaster, AL). Chloroform was obtained from Fisher Scientific. The 1,1′-dioctadecyl-3,3,3′,3′ tetramethylindotricarbocyanine iodide (DiR) was purchased from AAT Bioquest Inc. HEPES was purchased from Affymetrix, Inc. TiO2 beads were obtained from GL Sciences Inc. The 1,5-InsP8 and 1,5-PCP-InsP8 were synthesized as previously described.34 (link),46 The synthesis of 5-FAM-InsP7 began with a fluorenylmethyl derivative of InsP6 in which the only non-protected phosphate is at the 5-position.22 (link),48 The latter was phosphitylated with phosphoamidate and oxidized with meta-chloroperoxybenzoic acid following a strategy that proceeds via P(iii)–(v) intermediates.46 ,47 Both reactions proceeded with a yield of 50% underlining the efficiency of the approach. Next, the alkyne modified product was used in an azide–alkyne Huisgen cycloaddition under copper catalysis resulting in the fluorescein modified FAM-5-InsP7 product with a yield of 93% (see ESI). The final preparation of 5-FAM-InsP7 was validated by PAGE62 (link) and by NMR (see ESI). The origin of the WT and PPIP5K KO HCT116 cells has been described previously.63 (link) The culture medium for these cells was DMEM/F12 plus 10% FBS (Germini Bio-Product) and 100 U mL–1 penicillin–streptomycin (ThermoFisher Scientific) at 37 °C with 5% CO2.

Synthesis and Characterization of Polymeric Nanoparticles

N-t-butoxycarbonyl-phenylalanine (Boc-Phe) was provided by GL Biochem Co, Ltd (Shanghai, People’s Republic of China), and 1,1′-dioctadecyl-3,3,3′,3′-tetramethylindotricarbocyanine iodide (DiR) was supplied by AAT Bioquest, Inc (Sunnyvale, CA, USA), respectively. Acetonitrile and methanol were both of HPLC grade and were purchased from Oceanpak Alexative Chemical (Gothenburg, Sweden). All other chemicals were reagent grade and were used as received. Purified water used throughout the study was provided by Wahaha Group Co, Ltd, (Hangzhou, People’s Republic of China).
