Western Blot Analysis of EV and Cell Proteins

Cells and EVs were lysed in RIPA buffer (Merck). Proteins (20 μg for cells and 6 µg for EVs) were electrophoresed on SDS-PAGE. Western blotting analyses were performed using the following antibodies: mouse monoclonal anti-human CD81 (Santa-Cruz Biotechnology; SC-166029), mouse monoclonal anti-human CD63 (Santa-Cruz Biotechnology; SC-5275), rabbit monoclonal anti-human CD9 (abcam; ab92726), rabbit monoclonal anti-human Ang2 (abcam; ab155106), rabbit polyclonal anti-human p-Akt (Cell Signaling technology; #9271), rabbit polyclonal anti-human p-ERK1/2 (Cell Signaling technology; #9101), mouse monoclonal anti-human PI3K (abcam; ab86714), and mouse monoclonal anti-human beta-Actin (Cell Signaling technology; #3700). For the expression of ANG2, p-Akt, p-ERK1/2 and PI3K, the cells were cocultured with 10 µg/mL EVs for 24 h prior to analysis. Densitometry readings were measured using FUSION FX software and were normalized to the corresponding β-actin. The original WB can be found in Figures S1 and S2.
