Lineage Tracing of Aldh1l1+ Cells

All animal experiments were performed in accordance with the guidelines of the Institutional Animal Care and Use Committee of University (Approval number: 2018–059). The protocol was reviewed and approved by the Ethics Committee on Laboratory Animal Care. The mice were housed in rooms with controlled 12 hr light/dark cycles, temperature, and humidity, and food and water were provided ad libitum. Eight- to 10-week-old C57BL/6 mice weighing 22–26 g were obtained from the Vital River Laboratory Animal Technological Company (Beijing, China). Aldh1l1-CreERT2 transgenic mice (Stock number #029655), Rpl22 lsl-HA (Ribotag) mice (Stock number #011029), and Rosa26lsl-YFP mice (Stock number #006148) were obtained from The Jackson Laboratory (Bar Harbor, ME, USA). Aldh1l1-CreERT2 mice were used for breeding to the Rpl22lsl-HA mice or Rosa26lsl-YFP mice. Eight- to 10-week-old Aldh1l1-CreERT2;Rpl22lsl-HA and Aldh1l1-CreERT2;Rosa26lsl-YFP mice were used for lineage-tracing experiments.

Lgr5-Ezh2-Eed Mouse Model Generation

Lgr5EGFP-IRES-CreERT (Barker et al., 2007 (link)) and ROSA26lsl-yfp mice were purchased from The Jackson Laboratories, and Ezh2+/Fl mice (Su et al., 2003 (link)) from the Mutant Mouse Regional Resource Centers. VillinCreER-T2 (el Marjou et al., 2004 (link)) and Eedfl/fl mice (Xie et al., 2014 ) were generous gifts from S. Robine (Institut Pasteur, France) and S. Orkin (Boston Children’s Hospital), respectively. Mice were housed in a colony maintained at 23 ± 1°C, 55 ± 15% humidity, and 12 hr light/dark cycles, and handled according to protocols approved by the Animal Care and Use Committee of the Dana-Farber Cancer Institute. Mice were genotyped by PCR before weaning and again at the time of experiments.
