Detailed Analytical Protocol for Bone and Metabolite Studies

Formic acid (HPLC grade), 2-chloro-L-phenylalanine (as an internal standard), xanthine, L-phenylalanine, D9-phenylalanine, choline, hypoxanthine, L-tryptophan, succinic acid, and L-tyrosine standards were purchased from Sigma (Saint Louis, MO, USA). The animal treatment used corn oil (GLBIO, Montclair, CA, USA) and testosterone (GLBIO, Montclair, CA, USA). An 80% paraformaldehyde stationary liquid (Labgic Technology, Beijing, China) was applied for sample preservation.
For the decalcification of bone tissue, EDTA-2Na and sodium hydroxide were purchased from Sinopharm Chemical Reagent Co., Ltd., Shanghai, China., We applied the same pretreatment process for Hematoxylin-eosin (HE) and tartrate-resistant acid phosphatase (TRAP) staining. Anhydrous ethanol was purchased from Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China). An environmentally friendly dewaxing agent (alkane mixture), a transparent agent (alkane mixture, xylene-free), and other dye tools were purchased from Tonsen tech, Wuhan, China. For further staining, the HE dye kit was purchased from Biossci (Beijing, China), model BP092. The TRAP dye kit was purchased from Biossci (Beijing, China), model BP088.

Quantifying Reactive Species in PRO Degradation

A series of quenching, ESR and probe tests were conducted to identify the presence of multiple reactive species (e.g., h+, 1O2) and quantify their contributions to the PRO degradation during the EMF tests. For the quenching tests, a 4 mM portion of EDTA-2Na (Sinopharm Chemical Reagent Co., Ltd., China) and a 16 mM portion of FFA (Shanghai Macklin Biochemical Co., Ltd., China) were separately added to the feed solution (20 mg L−1 PRO, 10 mM Na2SO4) to quench the possibly generated h+ and 1O2, respectively. For the ESR tests, a 100 mM portion of TEMP (Meryer Co., LTD., China)66 (link) was added as the trapping agent of 1O2 to a 10 mM Na2SO4 solution, which was then used as the feed for the EMF experiment. The effluent was analyzed using an X-band ESR spectrometer (JES-FA200, JEOL, Japan). The adopted recording parameters were 1 mW for microwave power, 15 mT for sweep width, 0.1 mT for modulation amplitude, and 120 s for sweep time. Probe tests were conducted to identify the presence of ·OH7. A trace amount of TA (100 μM) was added to a 10-mM Na2SO4 solution to capture ·OH, which was then changed to hTA.

Removal of PAHs from Contaminated Sites

Comprehensive Bone Tissue Analysis Protocol

(Shanghai, China). The environmentally friendly dewaxing agent, transparent agent and sealed tablet were purchased from Tonsen tech, Wuhan, China. The histopathological staining kits were purchased from Biossci (Beijing, China) for HE staining (model BP092), TRAP staining (model BP088), and ALP (Model BP090).
