Intravital Imaging of Bone Resorption

To perform intravital imaging, lipopolysaccharide (LPS) (10 mg/kg body weight; Sigma-Aldrich) dissolved in phosphate-buffered saline (PBS) was injected into the calvarial periosteum of mice under isoflurane anesthesia. To perform RNA sequencing (RNA-Seq), reverse transcription-quantitative polymerase chain reaction (RT-qPCR), and flow cytometry analyses, LPS (20 mg/kg body weight) dissolved in PBS was injected into the calvarial periosteum. From the day of LPS injection, the JAK inhibitor ABT-317, which was provided by AbbVie Bioresearch Center, at a dose of 60 mg/kg body weight dissolved in 0.5% hydroxypropyl methylcellulose (Alfa Aeser) and 0.02% Tween 80 (Sigma-Aldrich) in sterile water was orally administered twice daily for 5 days. To visualize osteoclastic bone resorption using intravital bone imaging, a pH-activatable fluorescent probe for osteoclast activity sensing (pHocas)-3 dissolved in PBS was subcutaneously injected at a dose of 6 mg/kg body weight daily into TRAP-tdTomato mice, beginning 3 days before imaging. The C–C motif chemokine receptor 1 (CCR1) antagonist, J-113863 (MedChemExpress, #HY-103360) (3 mg/kg body weight) dissolved in 10% dimethyl sulfoxide (Sigma-Aldrich), 40% polyethylene glycol 300 (Wako), 5% Tween 80 (Sigma-Aldrich), and 45% PBS, was injected intraperitoneally once daily for 5 days.

Formulation of APX and CLOP Tablets

A pharmaceutical grade APX and CLOP hydrogen sulfate salt were received as gifts from Pharmacare PLC (Palestine). Tablet excipients including hydroxypropyl methylcellulose, 28–30 % methoxyl, 7–12 % hydroxypropyl, viscosity (2 % aq. soln., 20 °C) 7500–14000 mPa.s (HPMC) and hydroxypropyl cellulose (HPC) were purchased from Alfa Aesar (by Thermos Fisher Scientific, United Kingdom). Methocel, sodium laurilsulfate (SLS), spray-dried lactose, mannitol, microcrystalline cellulose 112 (MCC 112), colloidal silica, sodium stearyl fumarate (SSF), magnesium stearate (Mg. stearate), and anhydrous ethanol were donated from Pharmacare PLC (Palestine). For buffer preparation, sodium hydroxide pellets, sodium phosphate tribasic dodecahydrate, 98 %, and hydrochloric acid were analytical grades and purchased from Carlo Erba reagents, Acros organics, and Merck Serono respectively. HPLC grade solvents for analysis including; acetonitrile (ACN), trifluoroacetic acid (TFA), and triethylamine (TEA) were purchased from Merck (Merck Serono Amman, Jordan). Water was obtained by filtration using a cellulose nitrate filter (0.45) micron manufactured by Sartorius stedim biotech company (Jordan). Translucent hard gelatin shell capsules (0 size) were donated from Jerusalem Pharmaceuticals Co. Ltd. (Palestine).

Poloxamer-188 Formulation for Diltiazem Delivery

Comprehensive Formulation Development Protocol

NAC crystals and NAC powder (80 mesh) were used as provided by Nestle. Candelilla wax (CanW, melting point 68–72 °C), carnauba wax (CW, melting point 82 °C, no. 1 yellow, refined), sorbitan monooleate or Span 80 (saponification value: 145 to 160), formic acid (FA 98%) and acetonitrile (HPLC grade, ≥99.9%) were purchased from Sigma-Aldrich. Rice bran wax (RBW, melting point 79–85 °C) was purchased from Nutley's Kitchen Gardens. Beeswax (BW, DR-101, melting point 62–65 °C) was purchased from Strahl & Pitsch, Inc. Xanthan gum was purchased from TIC Gums. Carboxymethyl cellulose (CMC) was purchased from Sigma (low viscosity, 50–200 mPa s, 4% in water). Dioctyl sulfosuccinate sodium salt or AOT 96% was obtained from VWR. Hydroxypropyl methylcellulose (HPMC) purchased from Fisher (40–60 mPa s, 2% in water), sorbitan monostearate or Span 60 purchased from TCI America (saponification value: 145 to 156), and hexanes were purchased from Fisher (≥98.5%, Certified ACS, Fisher). Corn oil was purchased from a local market. Milli-Q and DI water were used for analyses and experiments, respectively. All chemicals were used as received without any further purification.

Formulation and Characterization of GMO-Based Lipid Nanoparticles

Povidone K 90 (polyvinyl pyrrolidone) was purchased from BASF Chemicals. Polyethylene glycols (PEG), Eudragit E100, Eudragit L100, Eudragit S100, hydroxypropyl methyl cellulose (HPMC), mineral oil, and olive oil purchased from Fisher Scientific. Deionized water was used for all the experiments.
