Characterization of Ivy Leaf Extract

Ivy leaf dry extract EA 575® (DER 5-7.5:1, 30% ethanol; batch number 14B0310) was received from Engelhard Arzneimittel (Niederdorfelden, Germany) and is well characterised by 17 ingredients from the natural product classes of flavonoids, saponins, and dicaffeoylquinic acids identified via LC-MS analysis [7 (link)]. All reagents for luciferase assays were received from Promega (Mannheim, Germany) if not stated otherwise. Coelenterazine h was obtained from Prolume (Pinetop-Lakeside, AZ, USA). Adenosine, BAY 60-6583, forskolin, 5′-N-EthylcarboxamidoAdenosine (NECA), and SCH 442416 were obtained from Sigma-Aldrich (Crailsheim, Germany). TNFα was received from Merck (Darmstadt, Germany). PSB-603 and CGS 21680 were obtained from Biomol (Hamburg, Germany).

(2009) and Valant et al. (2014), respectively. 2‐Amino‐6‐[[2‐(4‐chlorophenyl)‐1,3‐thiazol‐4‐yl]methylsulfanyl]‐4‐[4‐(2‐hydroxyethoxy)phenyl]pyridine‐3,5‐dicarbonitrile (capadenoson) was purchased from Haoyuan Chemexpress (Cat# HY‐14917; Shanghai, China).
Fentanyl citrate was purchased from Jansen‐Cilac Ltd, UK. Medetomidine (Domitor), meloxicam, and atipamezole hydrochloride (Antisedan) were purchased from Pfizer, UK. Buprenorphine (Vetergesic) and pentobarbitone (Euthatal) were purchased from Alstoe Animal Health, UK. Tween and propylene glycol were purchased from Sigma‐Aldrich, UK.

Adenosine A1 Receptor Binding Assay

BCA (bicinchoninic acid) protein assay kit was obtained from Pierce Chemical Company (Rockford, IL). LUF5834 (an hA1R partial agonist) was synthesized in our laboratory as described previously [27 (link)]. Chinese hamster ovary (CHO) cells stably expressing the hA1R were obtained from Prof. Steve Hill (University of Nottingham, UK). All other chemicals were of analytical grade and obtained from standard commercial sources.
