Otic Vesicle Volume Measurement

A subset of the larvae that were injected with S-MOs or mcrs1 mRNA and processed for ISH were embedded in a gelatin-based medium (0.5% gelatin, 30% bovine serum albumin, 20% sucrose, hardened with glutaraldehyde [75 μl/ml]), and vibratome sectioned at 50 μm in the transverse plane. Serial section images were collected, and the outer edge of the otic vesicle and its inner lumen were traced on each side of the head (injected and control) in every section, and the areas of the entire otic vesicle (total) and of the lumen calculated using Olympus cellSens Standard software (version 1.17) and converted to volume by multiplying by section thickness. The volume of the otic vesicle tissue was calculated by subtracting lumen volume from total otic vesicle volume. Volumes were compared between control and injected sides by a two-tailed, paired t-test (p < 0.05).

Histological Tissue Analysis in Xenograft Tumor Study

The xenograft tumor tissue, liver, and kidney excised from each sacrificed mouse in each group were fixed in a 10% formalin solution and processed for paraffin embedding. The paraffin-embedded tissue cassettes were mounted onto a rotary microtome, sectioned to an approximate thickness of 5 µm, and stained with hematoxylin and eosin (H&E). The sections were analyzed using a fluorescence microscope (BX50, Olympus, Tokyo, Japan) equipped with a digital camera (DP74, Olympus).
