Azide-Induced Apoptosis in A549 Cells

A549 cells were plated on glass coverslips and then treated with indicated amounts of azide in the presence or absence of 25 μM cobinamide for 24 hours. The cells were fixed in 3.7% paraformaldehyde, permeabilized with 1% Triton-X-100, and incubated with a cleaved caspase-3-specific antibody (1:100 dilution) (Cell Signaling) overnight at 4°C. After incubation with a fluorescein isothiocyanate-conjugated secondary antibody (1:100 dilution) (Jackson ImmunoResearch, West Grove, PA), nuclei were stained using Hoechst 33342 dye. Immunofluorescence images were recorded using a Keyence BZ-X700 fluorescence microscope.

Apoptosis Quantification in Mouse Cecum
