KORD Actuator Salvinorin B Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
The KORD actuator salvinorin B (SALB; Cayman Chemical, Ann Arbor, MI) was prepared as described previously (Tuscher et al., 2018 (link)). SALB was dissolved in 100% DMSO (Fisher Scientific, Pittsburg, PA) to a concentration of 10 mg/mL, aliquoted, and stored at −20 °C. On the day of injection, an aliquot was thawed, briefly warmed to 40 °C, and vortexed to ensure that the compound was fully dissolved. SALB was injected intraperitoneally (i.p.) at a dose of 10 mg/kg. Post-training i.p. injection of this SALB dose impairs object placement memory consolidation in ovariectomized mice expressing the KORD in the dorsal hippocampus (Tuscher et al., 2018 (link)). Mice were weighed 1–2 days prior to behavioral testing to avoid additional stress on the testing day.

Inhibition of AgRP Neurons in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Peripubertal mice harboring at least one copy of an AgRP-Cre transgene were anesthetized with a ketamine/xylazine cocktail and injected bilaterally into the ARC (Bregma distance – X: ± 0.25; Y: −1.4; Z: −5.8) with 300nL of inhibitory DREADD virus (rAAV9-hSyn-dF-HA-KORD-IRES-mCitrine, 5x1012 genome copies (GC) per ml) (UNC Vector Core). Mice were allowed to recover for approximately ten days and then injected with salvinorin B (Cayman Chemical) or DMSO vehicle control (5mg/kg s.c. daily) for inhibitory experiments, for ten consecutive days.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
