Antibody Characterization in Cell Research

Indirect ELISA for GAA Antibody Detection

An indirect ELISA was used to detect GAA specific antibodies. Plates were coated with rGAA (Myozyme, Genzyme Corporation) overnight, rinsed, and blocked with 5% milk in PBS. Samples in serial dilutions, controls, and blanks were added and incubated for 2 h at 37 °C. HRP conjugated donkey anti-rat IgG (1:5000, r712–035-150; Jackson Immuno Research) followed by Streptavidin/HRP (1:1000, P 0397; DakoCytomation) was added and 3,3′,5,5′- Tetramethylbenzidine (TMB, BD Biosciences) was used as substrate. Reactions were stopped with 2 N H2SO4 and reading was determined at 450 nm. Positivity was determined with control sera collected prior to the injections using a cut-off value of 0.200 OD.

Immunohistochemical Analysis of Germinal Centers

To examine GC architecture, spleens were removed from mice and fixed in 10% formalin (Sigma-Aldrich). For immunostaining, paraffin sections where deparaffined, hydrated and treated with antigen retrieval buffer (Dako). The sections then were permeabilized by 4 min treatment with Triton X-100 (0.1% in PBS) and incubated for 90 min with blocking buffer (Dako) followed by an overnight incubation with primary antibodies: rat anti-mouse B220 and hamster anti-mouse CD3e (1/1000, BD Pharmingen). For immunohistochemistry, antibodies were detected with AP-conjugated goat anti-Armenian hamster IgG or HRP-conjugated donkey anti-rat IgG (1/10000, Jackson ImmunoResearch Laboratories). HRP was reacted with DAB (Peroxidase Substrate Kit; Vector), and alkaline phosphatase with Fast Blue/Napthol AS-MX (Sigma-Aldrich). Levamisole (Sigma) was used to block endogenous alkaline phosphatase activity and slides were mounted in Crystal Mount (Electron Microscopy Sciences). Sections were viewed under a Nikon Microphot FXA light microscope and photographs were taken with a Spot Insight Camera, using 10× and 40× objectives, then analyzed using Spot Advanced software (Diagnostic Instruments) and ImageJ (NIH).

Immunophenotyping of Mouse Colonic Tissues

Mouse colonic tissues were harvested, swiss-rolled, and fixed with 4% paraformaldehyde (Sigma, St. Louis, MO) overnight. Fixed tissues were then embedded in OCT compound (Sakura, Tokyo, Japan) and kept at −80°C. Frozen tissue sections (7 μm) were first blocked for 30 min at room temperature with Blocking Reagent (PerkinElmer, Waltham, MA) and then incubated overnight at 4°C with primary antibodies as described below. Following overnight incubation, sections were washed and then incubated with HRP-conjugated secondary antibody for 1h at room temperature. After washing, sections were treated with tyramide-fluorescein (PerkinElmer), followed by counterstaining with DAPI (Sigma). All stained sections were analyzed by confocal microscope using either the 10× or the 20× objective (TCS SP2, Leica, Wetzlar, Germany). Images were processed with Adobe Photoshop software. The following antibodies were used: anti-CD11c and anti-F4/80 from BD Bioscience (Franklin Lakes, NJ), anti-CD3, anti-NKp46, and anti-B220 from eBioscience (San Diego, CA), HRP-conjugated donkey anti-rat IgG, and HRP-conjugated donkey anti-Armenian hamster IgG (H+L) from Jackson Immunoresearch (West Grove, PA).
