ADSCs were cultured with normal glucose (5.5 mM) or high glucose (25 mM; Invitrogen, CA, USA) medium for 6, 12, or 24 h. 3-Methyladenine (3-MA, 5 mM, Sigma, St. Louis, MO, USA) or rapamycin (100 nM, Aladdin, Shanghai, China) was employed to pretreat cells for 24 h to inhibit or induce autophagy in ADSCs, respectively. The cells were cultured in Dulbecco's modified Eagle's medium (DMEM) containing normal glucose (NG, 5.5 mM) or HG (25 mM), from Invitrogen; Thermo Fisher Scientific Inc., (Waltham, MA, USA), supplemented with 10% fetal bovine serum (Transgen Biotech, Beijing, China, http://www.transgen.com.cn/) and grown in a humidified incubator at 37˚C, in an environment containing 5% CO 2 .
