EN was purchased from Shanghai CP Guojian Pharmaceutical Co., Ltd. (Shanghai, China). HA (molecular weight 300kDa) was obtained from Freda Biochem Co., Ltd. (Shandong, China). Methacrylic anhydride (MA), 2-Hydroxy-4′-(2-hydroxyethoxy)-2-methylpropiophenone (I 2959), N,N′-methylenebisacrylamide (MBA) and Complete Freund’s Adjuvant (CFA) were obtained from SIGMA-ALDRICH (Merk, Darmstadt, Germany). Sulforhodamine B (SRB) was purchased from Yuanye Biotechnology Co., Ltd. (Shanghai, China). Trypan blue was purchased from Beyotime Biotechnology (Shanghai, China). TNF-α ELISA kit was purchased from Solarbio Life Science (Beijing, China) and IL-6 ELISA kit was purchased from MultiSciences (Hangzhou, China). All chemicals for preparation and analysis were obtained from the commercial source and used as received. Ultrapure water (18 MΩ cm; GenPure, Thermo Fisher Scientific, Waltham, MA, USA) was used for all solution preparations.

Collagen (collagen from chicken, type II) and tannic acid (TA) were obtained from Macklin Biochemical Technology Co., Ltd. (Shanghai, China). Triton X-100 was acquired from Sigma-Aldrich Inc. (Louis, MO, USA). An LDH kit was obtained from CAYMAN chemical company (Ann Arbor, MI, USA). Commercial adhesive (cyanoacrylate) was obtained from 3M (Japan). Isopropanol was obtained from Sinopharm (Beijing, China). Phosphate buffered saline (PBS) was obtained from Pricella (Wuhan, China). A TNF-α Elisa kit was obtained from Solarbio (Beijing, China). The TNF-α ELISA kit (Solarbio Life Science, Beijing) was used to detect TNF-α. The collected rat blood was placed at room temperature for 2 h and centrifuged at 1000 × g for 20 min before the supernatant was taken. After that, serum TNF-α concentration was detected by the TNF-α kit, and the detection steps were carried out according to the instructions. IL-1β, IL-6, Caspase-3, Caspase-9, LC3-II, ULK1, creatine kinase (CK), LDH, and ET-1 were detected using corresponding kits, and the operation was the same as above.

Take 50 μL of pre-treated rat serum and cell supernatant, add to ELISA microtiter wells and incubate for 1 h, then react with HRP for 30 min, after three washes with PBS for 5 min each time, and add the substrate TMB to develop the color for 15 min. TMB is converted to blue color catalyzed by the HRP enzyme, and converted to the final yellow color in the presence of acid. The absorbance (OD value) was measured at 450 nm with an enzyme marker and the concentration was calculated by standard curve.
