B was purchased from Baoji Guokang Bio-Technology Co.,Ltd (China). Chitosan (20 KDa), 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) (MTT) and acetic acid were obtained from Merck company (Germany). Also, culture medium RPMI-1640, Fetal Bovine Serum (FBS) and Penicillin/Streptomycin antibiotics were supplied by Gibco Company (USA). Furthermore, Tripolyphosphate (TPP), DMSO and Schneider culture medium were prepared form Sigma-Aldrich (USA). Iranian strain of L. major (MRHO/IR/75/ER) was supplied by Pasteur Institute of Iran. All other materials were of analytical grade. Double distilled water was used throughout the study.

Cultivation of Leishmania braziliensis

The strain of L. (V.) braziliensis (MHOM-BR-94-H3227) was grown in Schneider culture medium (Sigma Chem. Co., St. Louis, Mo, USA), containing 10% heat-inactivated foetal bovine serum (Sigma), 200 U penicillin/mL, and 200 mg streptomycin/μL (Sigma). The strain of L. braziliensis (MHOM-BR-94-H3227) was isolated from a patient with cutaneous leishmaniasis and characterized by polymerase chain technique and by monoclonal antibodies [28 (link)].

Culturing and Preserving Leishmania amazonensis Strains

The amastigote cultures of L. amazonesis MHOM/BR/pH8 strain were kept under cryopreservation until transferred to NNN medium (Novy–MacNeal–Nicolle) and cultured at 26 °C until the parasites reached the log growth phase. Then, the suspension was transferred to Schneider culture medium (Sigma-Aldrich, St. Louis, MO, USA), supplemented with 10% inactivated fetal bovine serum and 0.2% gentamicin sulfate, at 26 °C, so that the parasites returned to the log phase of growth. After that stage, the promastigote forms were incubated at 37 °C, producing a growth curve and the formation of the axenic amastigote strains [79 (link)].
The promastigote forms of L. amazonensis MHOM/BR/pH8 strain were preserved in Schneider medium (Sigma-Aldrich, St Louis, MO, USA) supplemented with 20% fetal bovine serum (FBS) and 1% streptomycin/penicillin, at 26 °C, in a B.O.D incubator—J. Prolab, São José dos Pinhais, Brasil, model JP. 100 (LBCM/CPAM). The promastigote forms were used in the exponential growth phase in all stages of the experiment [80 (link)].

Cultivation of Leishmania amazonensis Promastigotes

Leishmania amazonensis MHOM/Br/00/BA125 strain was used for DCs infection. L. amazonensis promastigotes were maintained in Schneider culture medium (sigma®) supplemented with 10% fetal calf serum (sigma®), 2 mm L-glutamine, 100 μg/ml penicillin and 100 μg/ml streptomycin (sigma®) in an incubator at 24°c. During the culture period, promastigotes were counted in the Neubauer chamber with a 100x dilution in saline in order to measure parasite growth dynamics. In all experiments, stationary phase parasites were used which are enriched by infective stage of parasite, called metacyclic promastigotes.

Lipid-based Formulations of Amphotericin B

Amphotericin B and AmBisome were gift from GeMePe, Argentina. Soybean phosphatidylcholine (SPC) (phospholipon 90 G, purity >90%) was a gift from Phospholipid/Natterman, Germany. Disodium succinate was from Baker Analyzed, USA. Modified Eagle's medium (MEM) was from Gibco, Argentina. Fetal calf serum (FCS), antibiotic/antimycotic solution (penicillin 10000 UI/ml, streptomycin sulphate 10 mg/ml, amphotericin B 25 g/ml), glutamine and trypsin/EDTA were from PAA Laboratories GmbH, Austria. Methanol, chloroform, hydrochloric acid and perchloric acid were from Anedra, Argentina.
