Quantitative LC-MS/MS Analysis of MTK

The sodium salt of MTK was obtained from KyongBo Pharmaceutical Co. Ltd. (Asan, South Korea). MTK and zafirlukast analytical standards were used as internal standards for LC-MS/MS. Polysorbate 80 (P80), tyloxapol, polyethylene glycol 4000 (PEG 4000), sodium carboxymethylcellulose (Na CMC), polyvinylpyrrolidone (PVP K17), acetic acid, sodium acetate, lactic acid, sodium lactate, and phosphate-buffered saline tablets were purchased from Sigma Chemical Co. (St. Louis, MO, USA). Polyoxyl 15 hydroxy stearate (Kolliphor HS15), polyoxyl-35 castor oil (Kolliphor EL), and polyoxyl 40 hydrogenated castor oil (Kolliphor RH40) were provided by BASF (Ludwigshafen, Germany). HPLC-grade acetonitrile and methanol were purchased from J. T. Baker (Phillipsburg, NJ, USA). All other chemicals were of analytical grade and used without further refinement.

Analytical grade chemicals, reagents, and internally prepared double distilled water were used in this research.

Optimized siRNA Delivery System

RH40, Polyoxyl 35 castor oil (Cremophor®EL) were purchased from BASF (Ludwigshafen, Germany). Glycerol and Propylene glycol were provided by Fagron Nordic A/S (Rotterdam, The Netherlands). All other reagents were of pharmaceutical grade. Reagents for cell culture were provided by Gibco (Montpellier, France).
A 21-base pair siRNA oligonucleoside duplex for silencing B2M gene was synthesized by the help of ‘siRNA Explorer’ and dissolved in diethylpyrocarbonate (DEPC)-treated water before use. The gene sequence was confirmed by HPLC-MS and gel electrophoresis comprehensively. The gene silencing efficiency was verified in vitro. Jet-PEI was provided by Polyplus Company. Lipo2000 was provided by Invitrogen Company.
NIH female mice (8 weeks, 20 ∼ 28 g) in estrous phase were provided by Guangdong Medical Experimental Animal Center (China). The study protocol was reviewed and approved by the Animal Welfare and Ethics Committee of Tsinghua University.

Medium-chain triglycerides (Miglyol 810N) and tricaprylin (Captex 8000) were obtained from IOI Oleo GmbH (Witten, Germany) and Abitec Corporation (Columbus, OH, USA), respectively. Sodium metabisulfite was purchased from Daejung Chemicals & Metals (Siheung-si, Korea). HPLC-grade methanol was purchased from J.T. Baker (Phillipsburg, NJ, USA). All other reagents or chemicals were of analytical grade and were employed without further purification.

Amoxicillin Trihydrate Formulation and Characterization

Sodium hydroxide (GR grade), sodium lauryl sulfate (GR grade), sodium dihydrogen phosphate (GR grade) and phosphoric acid (GR grade) and glacial acetic acid (GR grade) were purchased from Sinopharm Chemical Reagent Co., Ltd (Shanghai, China). Methanol (HPLC grade) and acetonitrile (HPLC grade) were purchased from Merck Pvt. Ltd. (Shanghai, China). Polyoxyl 35 castor oil was purchased from BASF (Germany). Hydroxypropyl methyl cellulose (HPMC E5) and Opadry® were purchased from Colorcon (Shanghai, China).
