Human VSMCs (T/G HA-VSMC) purchased from Procell Life Science & Technology (Wuhan, China) were cultured in Ham's F-12K (Procell Life Science & Technology) with 10% FBS, 100 μg/mL streptomycin, and 100 U/mL penicillin in an incubator containing 5% CO2 at 37°C.
For cell transfection, cells were inoculated into poly-D-lysine-coated 6-well plates at the concentration of 2 × 105. Then, the sequences of miR-21 and the negative control (miR-NC) designed by Sangon Biotech (Shanghai, China) were transfected into T/G HA-VSMC cells using Lipofectamine 3000 (Thermo Fisher Scientific, Waltham, MA, USA).

Culturing Human Vascular and Renal Cells

Culturing Human Aortic VSMCs and HEK-293T Cells

Human aortic VSMCs were cultured in smooth muscle cell medium (SMCM; ScienCell Research Laboratories, Inc.) which contained basic medium, 2% fetal bovine serum, 1% smooth muscle cell growth supplement and 1% penicillin/streptomycin solution. A human embryonic kidney (HEK)-293T cell line was purchased from the Institute of Basic Medical Sciences, Chinese Academy of Medical Sciences (Beijing, China) and maintained in Dulbecco's modified Eagle's medium (Hyclone; Cytiva) containing 10% fetal calf serum (Gibco; Thermo Fisher Scientific, Inc.). Both cell lines were kept in a humidified atmosphere of 5% CO2 at 37˚C.
