CC-3156) supplemented with 10% fetal bovine serum (FBS; GIBCO, Grand Island, NY, USA), 100 units/ml penicillin (GIBCO), 100 μg/ml streptomycin (GIBCO), and endothelial growth supplement (BD Biosciences). Culturing Primary Human Endothelial Cells

Human umbilical vein endothelial cells (HUVECs) from single donors were purchased and cultured in endothelial cell growth medium (Cell Applications, San Diego, CA, USA). Early passage (1–3), single donor HUVECs were used for all experiments. Pulmonary Arterial Endothelial Cells (PAEC; passage 6 for Donor 1 and passage 7 for Donor 2) from donors without history of pulmonary hypertension were obtained from the Cleveland Clinic’s Biorepository [41 (link)]. PAEC were grown in MCDB105 medium (Sigma, Burlington, MA, USA) supplemented with 90 µg/mL heparin (Sigma, Burlington, MA), 90 µg/mL endothelial growth supplement (BD Biosciences, San Jose, CA, USA), 15 µg/mL glycine, 15 µg/mL KCl, 15% heat inactivated fetal bovine serum and normocin (Invitrogen, San Diego, CA, USA) as an antibacterial-antimycotic-antimycoplasmic agent.
