Cells were washed twice with PBS before being fixed in 4% paraformaldehyde for 20 min at room temperature, permeabilized in PBS with 0.2% Triton X-100 for 10 min, and blocked in PBS with 10% normal donkey serum for 30 min at room temperature. The cells were then incubated with diluted primary antibody in PBS with 10% donkey serum at 4°C overnight followed by incubation with the corresponding secondary antibody in the dark at room temperature for 40 min. The cells were washed with PBS three times between each step. The nuclei were counterstained with 4′,6-diamidino-2-phenylindole (DAPI) before visualization and image acquisition under a microscope.
