Quantifying Fc-mediated C1q Binding

The interaction of Fc variants with mouse C1q was measured by ELISA. MaxiSorp 384-well plates (Thermo Fisher Scientific) were directly coated with the anti-CD154 antibodies overnight at 4 °C. After blocking with TBS-T containing 0.5% BSA and 1 × Block Ace (DS Pharma Biomedical) for 7 h at 4 °C, 10% mouse plasma (Innovative Research, Novi, MI, USA) was added onto the plates and incubated overnight at 4 °C. At room temperature, the plates were incubated with a biotinylated anti-mouse C1q antibody (Hycult Biotech, Uden, The Netherlands) for 1 h. Pierce High Sensitivity Streptavidin-HRP (Thermo Fisher Scientific) was added to react for 1 h at room temperature. Washes in PBS-T (pH 7.4) were performed after each subsequent step. ABTS peroxidase substrate (SeraCare Life Sciences, Milford, MA, USA) was subsequently added, and the signal was measured by a plate reader at a wavelength of 405 nm.

Phlpp Inhibitor Stability Assay

To test the stability of Phlpp inhibitors in vitro, 8 ​μM of NSC117079 or NSC 45586 were added to culture medium (DMEM supplemented with 10% FBS) and mouse plasma (Innovative Research). Samples were collected in triplicate after incubation at 37 ​°C for 5, 15, and 30 ​min, and 1, 2, 4, 8, 12, 24, 48 and 72 ​h. Molecule concentrations were measured with LC/MS.

Sestrin 2, zonula occludens-1 (ZO-1), and glyceraldehyde-3-phosphate dehydrogenase (GAPDH) rabbit polyclonal primary antibodies were purchased from Proteintech (Rosemont, IL, USA). Horseradish peroxidase (HRP)-linked anti-rabbit secondary antibodies were purchased from Cell Signaling Technology (Danvers, MA, USA).

Quantification of Benzo[d]thiazole Compounds

LCMS-grade acetonitrile, methanol, isopropyl alcohol, water, and formic acid were obtained from Fisher Scientific (Fair Lawn, NJ, USA). Mouse plasma was purchased from Innovative Research, Inc. (Novi, MI, USA). MCI-77 [3-(2-(azepan-1-yl)ethyl)-6-benzylbenzo[d]thiazol-2-(3H)-one hydrochloride], the internal standard (IS) MCI-147 [3-(2-(azepan-1-yl)ethyl)-6-(3,4-dichlorobenzoyl)benzo[d]thiazol-2(3H)-one hydrochloride], and the parent compound CM-304 [3-(2-(azepan-1-yl) ethyl)-6-(3-fluoropropyl)benzo[d]thiazol-2(3H)-one hydrochloride] were synthesized in house. The purity of the compounds was confirmed by high-performance liquid chromatography, high-resolution mass spectrometry, and 1H and 13C nuclear magnetic resonance.

Plasma-Based Tumor Characterization

Mice were anesthetized via isoflurane for sample collection. Blood was collected via cardiac puncture and immediately centrifuged to separate and collect the plasma samples. Tumors were surgically removed promptly after mice were euthanized and then homogenized in 300 μL mouse plasma (Innovative Research) for 30 m using a cordless pestle motor (VWR). Cell matter was then removed via centrifugation. Mice were sacrificed at 5 min, 30 min, 2 h, 6 h, 12 h, 24 h, 48 h, and 72 h post injection.
