Alport Syndrome Mouse Model Treatment

Four groups of eight 129sv mice were treated: Wild type with 0.5% methylcellulose (SIGMA) as vehicle, Wild type with PAT-1251 (provided by PharmAkea, Inc.) at 30 mg/kg in 0.5% methocel, Alport with vehicle, and Alport with PAT-1251 at 30 mg/kg in 0.5% methylcellulose. The treatments were given once daily by gavage needle from 2-7 weeks of age.
Urine was collected weekly from 4-7 weeks. Albumin was measured using Molecular Innovations (Novi, MI) Mouse Albumin Antigen ELISA Kit Cat# MSAKT. The albumin levels were normalized to creatinine measured using BioAssay Systems (Hayward, CA) Creatinine Assay Kit (DICT-500) according to the manufacturer’s instructions. BUN measurements were determined with BioAssay Systems Urea Assay Kit (DIUR-100) according to the manufacturer’s instructions.

Dapagliflozin Attenuates Albuminuria in db/db Mice

Male db/db (Leprdb/db) and m/m mice (C57BL/KsJ mice) were obtained from Nanjing Biomedical Research Institute of Nanjing University. The mice were housed in the Animal Center of Peking University First Hospital with a 12-h light/dark cycle allowing free access to food and water when they were 3 weeks old. After 1 week (at the age of 4 weeks), db/db mice were randomly divided into two groups, with 7 mice for each group. For dapagliflozin-treated group, the dosage of dapagliflozin (AstraZeneca Pharmaceuticals LP) was 1 mg/kg/day and the solvent was 0.5% methylcellulose. The control groups received solvent as vehicle without active pharmacological effect.
24-h urine was collected to measure urinary albumin at the 8th and 14th week. Mice were placed in a 24-h metabolic cage for urine collection. Urinary albumin levels were measured by a mouse albumin ELISA kit (Bethyl Laboratories, Montgomery, TX, United States). Urinary creatinine levels were assayed by a Creatinine Assay Kit (DICT-500; BioAssay Systems, CA, United States). The mice were sacrificed at the age of 14 weeks. No adverse effect was found in dapagliflozin-treated db/db mice. All animal experiments were approved by the Laboratory Animal Ethics Committee of Peking University First Hospital (No. 202039).

Animal Model of Diabetic Nephropathy

The animal experiments were approved by the Institutional Animal Care and Use Committee of Jinling Hospital (Nanjing, China), in accordance with the approved guidelines of the Institutional Animal Care and Use Committee of Jinling Hospital. 7 weeks old male wild-type (wt) and leptin receptor-deficient (db/db) mice on the C57BLKS/J background were purchased from the Model Animal Research Center of Nanjing University (Nanjing, China). All animals were maintained in rooms with constant temperature and humidity under a 12-h light/dark cycle. Body weight and fasting blood glucose levels were monitored biweekly with a glucometer. Murine urinary albumin and creatinine levels were measured using an Albuwell M (1011, Exocell, Philadelphia, USA) and Creatinine Assay Kit (dict-500, Bioassay systems, CA, USA) according to the manufacturer’s instructions.

Renal Function Evaluation in Mice

Body weight and renal weight measurements of mice were accurate to 0.01 g with electronic scales. Kidney-to-body weight ratio was calculated as left or right kidney weight (g) divided by the body weight (kg). After anesthesia with avertin, heparin-anticoagulated blood of mice was collected from the angular vein. BUN, TCHO, and triglyceride levels in plasma were determined with a UniCel DxC 600 Chemistry Analyzer (Beckman Coulter). 24 hours’ urine was collected in the metabolic cage, and urinary albumin concentration was measured by a mouse albumin ELISA kit (Bethyl Laboratories). Urinary creatinine level was measured by a Creatinine Assay Kit (DICT-500; BioAssay Systems). uACR was calculated as urinary albumin (μg) divided by urinary creatinine (mg).
