Peptide Conjugation and Polyclonal Antibody Production

A 117 amino-acid peptide corresponding to RH5Nt (F1-K116) with an additional C-terminal cysteine was chemically synthesized (LifeTein). The peptide was dissolved in DMF and conjugated to maleimide-PEG2000-DSPE (Avanti Polar Lipids) at a 1:2 molar ratio overnight at room temperature with rotation. Polyclonal antisera were raised by immunizing rabbits six times with 100 μg of purified antigen in Freund's adjuvant in rabbits (Cambridge Research Biochemicals) in strict accordance with UK Home Office governmental regulations and the local Sanger Institute animal welfare ethical review board. Polyclonal antisera were similarly raised against full-length CyRPA and the P113 Y1-N653 fragment expressed in mammalian cells as rat Cd4d3+4-6His tagged proteins and purified using Ni2+-NTA chromatography. The sera were purified according to manufacturer's instructions on an ÄKTA system using separate Hi-Trap Protein G HP columns (GE Healthcare) for each rabbit serum. The fractions were eluted with 0.2 M glycine (pH 2.7) directly into a neutralization buffer (1 M Tris:Cl pH 9.0) and immediately dialysed into PBS.

Trastuzumab Conjugation with DSPE-PEG-Maleimide

Lyophilized Trastuzumab (MedChemExpress) was reconstituted in PBS. First, Trastuzumab was reacted with excess 2-iminothiolane (1:20 mol/mol) in an oxygen free reaction vessel filled with gaseous N2 for one hour. Then, the mixture was filtered using PD 10 desalting columns to remove excess 2-iminothiolane. Trastuzumab concentration was measured using Precision Red Advanced Protein Assay (Cytoskeleton, Inc., Denver, CO) and filtered samples were then incubated stirring overnight with 1,2-distearoyl-sn‑glycero-3-phosphoethanolamine-N-[maleimide(polyethylene glycol)−2000] (ammonium salt) (DSPE-PEG-2000) Maleimide (Avanti Polar Lipids, Birmingham, AL) in a 1:4 Trastuzumab:Lipid mol/mol ratio. Samples were collected after each reaction to measure thiol concentrations using Measure-iT™ Thiol Assay Kit (Sigma Aldrich) following manufacturer's protocol.

Lipid Nanoparticle Formulation Protocol

Lipid stock solutions were formulated in chloroform. 140 μL DOPC (10 mg/mL), 30 μL DSPE-PEG(2000) maleimide (5 mg/mL), 150 μL cholesterol (5 mg/mL), and 50 μL 18:1 PEG 2000-PE (5 mg/mL, all purchased from Avanti Polar Lipids) were combined to attain a DOPC:DSPE-PEG(2000) maleimide:cholesterol:PEG 2000-PE mass ratio of 55:5:30:10 and 2.5 mg total lipid. Chloroform was evaporated under a stream of nitrogen and residual solvent was removed under vacuum overnight.
