Kidney Protein Extraction and Western Blot Analysis
five volumes of homogenate buffer composed of 10 mM HEPES buffer, pH 7.9, 10 mM KCl,
0.1 mM egtazic acid, 1 mM dithiothreitol, and 0.5 mM phenylmethylsulfonyl fluoride.
The kidney homogenates were centrifuged at 3000 g for 15 min at 4°C,
and the supernatants were subsequently stored at -80°C. The protein extracts from the
kidney tissues were separated on 12% SDS-PAGE gels and transferred to polyvinylidene
difluoride membranes. Gapdh served as a loading control.
Bmp7, Igf2, Tgfβ,
Hgf, and Ctgf expression were evaluated using
rabbit polyclonal anti-Bmp7 (1:250; Bioworld Technology, USA),
anti-Hgf (1:500; Abcom, USA), anti-Tgfβ (1:250;
Bioworld Technology), anti-Ctgf (1:3000; Abcom), and
anti-Igf2 (1:150; Bioworld Technology) antibodies, which were
purified for immunogen affinity. The bands were quantified using the Image-Pro Plus
software (USA).
