Absolute Quantification of Leukocyte Subsets

Check if the same lab product or an alternative is used in the 5 most similar protocols
The collected leukocytes from peripheral blood, mesenteric LN, lung, and bone marrow were incubated with Fc-blocking TruStain FcXTM (anti-mouse CD16/32, BioLegend, San Diego, CA, USA) antibody, followed by Zombie aqua (live/dead indicator, BioLegend, San Diego, CA, USA). The cells were stained with fluorescence-labeled antibodies. The following antibodies were purchased from BioLegend (San Diego, CA, USA): BV650-CD45.2 (104), Alexa Flour 700-CD3 (500A2), Pacific Blue-CD4 (RM4-5), BV570-CD8 (53-6.7), BV711-CD19 (6D5), and FITC-Gr-1 (RB6-8C5). All samples were acquired on a Cytek Aurora flow cytometry (Cytek Biosciences, Fremont, CA, USA), and the data were analyzed with FlowJo software (BD Bioscience, Franklin Lakes, NJ, USA). The calculation of the absolute number of leukocytes was carried out using CountBrightTM Absolute Counting Beads (Invitrogen, Waltham, USA) and according to the recommended manufacturer manual, with modifications. To calculate the absolute count, the following equation was applied: Absolute count cells=Number of cell events×assigned beads count of the lotNumber of bead events×1000 µLThe vol of 2×106 cells

Isolation and Profiling of Intestinal Immune Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
After 4-week
exposure, the intestines of mice were collected and the intestinal
cells were isolated, washed, and diluted with phosphate-buffered saline
(PBS, pH = 7.4, Solarbio, China) containing 0.1% fetal bovine serum
(FBS, Gibco). Then, the cells (2 × 106) were blocked
with TruStain fcX TM (antimouse CD16/32, Biolegend Inc.) at 4 °C
for 15 min and stained with an array of fluorescent-labeled antibodies
(Biolegend Inc) at 4 °C for 30 min, and then, cells were identified
by flow cytometry (Attune NxT, Thermo Fisher Scientific). Details
of antibodies (Abs) used are provided in the Supporting Information.

Intestinal Cell Immunophenotyping

Check if the same lab product or an alternative is used in the 5 most similar protocols
The intestinal cells were collected
after 4-week exposure, then the cells (2 × 106) were
blocked with TruStain fcX TM (antimouse CD16/32, Biolegend Inc.) at
4 °C for 15 min and stained with FITC-conjugated antimouse MHC
II (M5/114.15.2) Ab (Biolegend Inc), APC/Cy7 conjugated antimouse
CD11c (N418) Ab (Biolegend Inc.), and propidium iodide (PI, Biorigin,
China). Dead cells were identified by flow cytometry (Attune NxT,
Thermo Fisher Scientific).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
