pDC Stimulation and Cytokine Assay

Isolated spleen or BM pDCs (100k) were resuspended in 100 μL of complete 1640 medium and then stimulated in a 96-well U-bottom plate with 1-μM ODN2216 or 10-μg/mL poly U (Invivogen, San Diego, CA, USA) supplemented with Lipofectamine 2000 (ThermoFisher Scientific) (1.5 μL: 1000 μL) for 48 h. The supernatants were collected and stored at −80 °C for enzyme-linked immunosorbent assay (ELISA). Each group consisted of at least 3 mice, and each experiment was repeated at least 3 times.

Investigating HIV Infection and Immune Response in Memory CD4 T Cells

Most experiments involved culture of uninfected or HIV-infected memory CD4 T cells in IL2 (20ng/ml) medium for up to 1 week, followed by data collection. Cells were cultured in 48-well plates (5x105 cells in 1ml medium) with appropriate reagents. The HIV antiviral inhibitor, Saquinavir, was acquired from NIHAIDS Reagent Program and used at 1–10μg/ml. The GrzB/caspase inhibitor, Z-IETD-FMK (Enzo Life Sciences, Farmingdale, NY USA), was used at 10μM. Experiments involving TLR stimulation of memory CD4 T cells utilized synthetic TLR ligands LPS (Sigma, St. Louis, MO USA), Pam3CSK4 (EMD Millipore, Billerica, MA USA), peptidoglycan, Poly(I:C), and Poly(U) (InvivoGen, San Diego, CA USA), and were used at 2–25μg/ml +/− CD3 activation (clone UCHT1 coated at 0.1μg/ml).

Immunological Response Activation Assay

Mycophenolic acid, the active moiety of MMF, was purchased from Tokyo Chemical Industry. PSL, HCQ, and TPCA-1 were bought from Sigma-Aldrich. Poly(U) (InvivoGen) was complexed with the DOTAP Liposomal Transfection Reagent (Roche) at a ratio of 1 μg of Poly(U) per 6 μl of DOTAP and was used for stimulation. R-848 (resiquimod) was acquired from Enzo Life Sciences. CpG-B ODN 1668 (5′-T*C*C*A*T*G*A*C*G*T*T*C*C*T*G*A*T*G*C*T-3′) and CpG-A ODN 2216 (5′-G*G*GGGACGATCGTCG*G*G*G*G*G-3′) were synthesized by Eurofins Genomics (an asterisk indicates a phosphorothioate linkage). The monoclonal antibody that specifically recognizes phosphorylated IRF5 (anti-phospho-IRF5 antibody) was generated by the antibody phage display method53 (link) following the immunization of rabbits with a synthetic peptide corresponding to the region around phosphorylated serine 446 (S446) of human IRF5 isoform B (equivalent to the S462 of human IRF5 isoform D).

Synthesis and Characterization of Novel Polymeric Agents

Imidazole-1-sulfonyl azide hydrochloride [25] , N-(2-hydroxyl propyl)-methacrylamid (HPMA) and DMAE-CI [26] were synthesized as previously described. Ovalbumin (OVA) and PolyU were purchased from In-vivoGen (Toulouse, France). Cy5-mRNA_luc was from Teu-bio (TRiLink biotechnologies, San Diego, USA). Agarose multi-purpose was purchased from Roche Molecular Biochemicals (Mannheim, Germany). 6× DNA Loading Dye was purchased from Fermentas (St. Leon-Roth, Germany). SYBR Safe DNA gel stain, Dulbecco's phosphate buffered saline (DPBS), RPMI-1640/IMDM medium, Opti-MEM, and heat inactivated fetal bovine serum (HI-FBS) were purchased from Life Technologies (Breda, the Netherlands).
