Purification of Commercial hCP Enzyme

The commercial hCP (Cabiochem) was further purified by gel filtration chromatography, using Superose 6 FPLC column (Pharmacia, Sweden). Salsolinol, azide, formate, mannitol, bathocuproine disulfonate, diethyldithio carbamic acid (DDC) and penicillamine (PA) were obtained from Sigma (St. Louis, MO, USA). Chelex 100 resin (sodium form) was obtained from Bio-Rad (Hercules, CA, USA). All solutions were treated with Chelex 100 resin to remove any traces of transition metal ions.

Quantifying NADPH Oxidase Activity

NADPH oxidase activities were assayed with lucigenin enhanced chemiluminescence method as previous described47 (link)48 (link)49 (link). Briefly, cells were suspended and lysed in NADPH lysis buffer (50 mM phosphate buffer, pH 7.0, 1 mM EGTA, 150 mM sucrose, and protease inhibitors). Total cell lysate containing 100 μg proteins were incubated with 10 μmol diethyldithiocarbamic acid (Sigma) at 37°C for 30 min. Then mixtures were treated with 5 μmol lucigenin (Sigma) and 0.1 μmol NADPH (Sigma) balanced with NADPH lysis buffer. After incubation for 10 min at 37°C in dark, the samples were detected with luminometer. Reactions were terminated with 10 μmol tiron (Sigma). The enzyme activity was expressed as relative light units/μg protein and relative fold changes were used to indicate the activity changes.

Fabrication of Biocidal Cu2O-Infused Nonwoven Textiles

Polypropylene (PP) nonwoven textiles (Kimtech Pure W4 Critical Task Dry Wipers) were purchased from Kimberly Clark. Within this fabric were holes that were approximately 1 mm in diameter (See Figure 1). Cu2O particles (Chem Copp HP III Type UltraFine-5) were purchased from the American Chemet Corporation. Metrics of the particle size and chemical composition are given in the Supporting Material, Tables S1 and S2. Diethyldithiocarbamic acid (ACS reagent grade Sodium diethyldithiocarbamate trihydrate) was purchased from Sigma-Aldrich (St. Louis, MO, USA). Bicinchoninic Acid (Bicinchoninic Acid Disodium Salt > 98% purity) was purchased from Tokyo Chemical Industry (Tokyo, Japan). One hundred percent ethanol (EtOH ACS grade) was purchased from VWR. Deionized (DI) water was from a Milli-Q Reference (MilliporeSigma) water purification system.

Superoxide Anion Production in Mouse Aortas

Superoxide anion production in mouse aortas were measured by lucigenin-enhanced chemiluminescence assay. Briefly, dissected mouse aortas were incubated in Krebs-HEPES solution containing (in mmol L−1): 99 NaCl, 4.7 KCl, 25 NaHCO3, 1 KH2PO4, 20 Na-HEPES, 1.2 MgSO4, 2.5 CaCl2 and 11 glucose, supplemented with diethyldithiocarbamic acid (1 mmol L−1; Sigma-Aldrich) and β-NADPH (0.1 mmol L−1; Sigma-Aldrich) at 37 °C for 45 min. The aorta were then transferred to vials containing lucigenin (10 μmol L−1; Sigma-Aldrich) in Krebs-HEPES solution [29 (link)]. Repeated measurements were recorded by the GloMax® 20/20 Luminometer (Madison, WI, USA) in 1 min intervals for 10 min. The amount of superoxide anion produced was presented as real numbers in relative light units per mg of dry tissue [30 (link)].

Hormone Application in Tissue Culture

Before making plates, appropriate amount of individual hormone solutions were added into the culture medium around 50 °C. Jasmonic acid (Sigma), (S)-(+)-Ibuprofen (Sigma), Paclobutrazol (Sigma), Gibberellic acid (Sigma), Ancymidol (Sigma) were dissolved in ethanol. Aminoethoxyvinyl glycine hydrochloride (Sigma), AgNO3 (Sinopharm Chemical Reagent Co., LTD), 2,3,5-triiodobenzoic acid (Sigma) and Diethyldithiocarbamic acid (Sigma) were dissolved in ddH2O.
