The cytotoxic effect of compounds on Vero cells was assessed using the Cell Counting Kit-8 (CCK-8, ZOMANBIO, Beijing, China) according to manufacturer's instructions. Briefly, cells were seeded onto a 96-well cell plate and cultured until they grew into a single layer. The culture medium was removed and washed with PBS twice. The medium was replaced with cell maintenance solution in the presence of serial dilutions of each compound. The plates were incubated for 24 h when the medium was replaced with fresh DMEM containing 10% CCK-8 solution and cultured for 1.5 h. Then the OD value for each well was detected at 450 nm on a microplate reader (BMG Labtech POLAR star Omega). The assay was repeated three times. Cytotoxicity was analyzed according to the following formula: Cytotoxicity (%) = [(Abs sample) – (Abs blank)]/[(Abs negative control) – (Abs blank)] × 100.
