Yeast Strain and Plasmid Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Yeast strains and plasmids are listed in Table S4 and Table S5, respectively. The mig1Δ mig2Δ and mig1Δ mig2Δ mig3Δ yeast strains were a generous gift from Hans Ronne (Swedish University of Agricultural Sciences). Fluorescently labelled Vps4 strains were a gift from David Teis (Innsbruck Medical University). Polyclonal antibodies raised against GFP (Urbanowski and Piper, 1999 (link)) and Rsp5 (Stamenova et al., 2004 (link)), and monoclonal glyceraldehyde 3-phosphate dehydrogenase (GAPDH; clone 6C5; Ambion) and carboxypeptidase Y (CPY; clone 10A5-B5; Molecular Probes) antibodies, were used for immunoblot analysis. These antibodies were routinely used at 1:1000 dilution.

Immunoblotting Protein Expression Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Whole-cell protein extracts were used for analysis, and immunoblotting was conducted as described previously 22 (link). Antibodies against CDK4 (clone D9G3E; Cell Signaling Technology, Danvers, MA), CyclinD1 (clone G124-326; BD Biosciences, Franklin Lakes, NJ), GAPDH (clone 6C5, Ambion), p16INK4a (clone G175-405, BD Biosciences) were used as probes, and horseradish peroxidase-conjugated anti-mouse or anti-rabbit immunoglobulins (Jackson Immunoresearch Laboratories, West Grove, PA) were employed as secondary antibodies. The LAS3000 charge-coupled device (CCD) imaging system (Fujifilm Co. Ltd., Tokyo, Japan) was employed for detection of proteins visualized by Lumi-light Plus Western blotting substrate (Roche, Basel, Switzerland).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
