Immunofluorescence Assay of hnRNPA1 and eIF3a

4.5x10 5 cells were plated 24 hr. before performing IFA. Cells were fixed in 4% paraformaldehyde solution and incubated for 10 min. at 37'C, followed by methanol fixation for 10 min. The cells were block using 6% BSA for 1 hr. at room temperature, followed by incubation with primary antibodies for 2-3 hr. The cells were then washed 3 times in 1X PBS/0.05% Tween 20, then incubated with secondary antibodies followed by PBS/0.05% Tween washes. IFA was performed using hnRNPA1 (Sigma, R4528; 1:500 dilution), eIF3a (Abcam, 3411S; 1:500 dilution) and Alexa Fluor 488-or 568-labeled donkey anti-rabbit or goat anti-mouse secondary antibodies (1:1,500 dilution, Life Technologies, A10042, A11029), and mounted with DAPI-containing VECTASHIELD mounting medium (Vector Labs). The images were obtained at the CNR Biological Imaging Facility, University of California at Berkeley.
