Bioactive Compounds Analysis in Calendula Officinalis

Chlorogenic acid, rutin, 3,4‐dicaffeoylquinic acid (DA), isorhamnetin 3‐O‐glucoside (IG), calenduloside E (CE), curcumin, nomifensine and MPTP (1‐methyl‐4‐phenyl‐1,2,3,6‐tetrahydropyridine) were purchased from Shanghai Yuanye Biotechnology Co. Ltd. MPP (dihydrochloride) was purchased from Sigma‐Aldrich. Acetonitrile and MS‐grade water were acquired from Tedia Company Inc. and Watsons Ltd., respectively. All other chemicals were of analytical grade. SH‐SY5Y cells were obtained from the American Type Culture Collection (ATCC). Fresh flower heads of C. officinalis were obtained from the Anguo medicine market, Hebei Province. One gram of plant material was extracted with 8 mL of 70% ethanol for 1 h using ultrasound, and, subsequently, the solution was concentrated to acquire the residue (111.4 mg).

HPLC Analysis of Phenolic Compounds

Acetonitrile and methanol (HPLC grade) were purchased from Merck (Darmstadt, Germany). Ultrapure water was made using a Direct-Q 8 UV-R water purification system (Millipore, Billerica, MA, USA). Ltd. (Shanghai, China).

Isolation and Characterization of Flavonoid Glycosides

Astragalin (kaempferol-3-O-glucoside) and isoquercitrin (quercetin-3-O-glucoside) were extracted and isolated from crabapple fruits (Malus “Winter Red”) using the reported methods, and kaempferol and quercetin were obtained by hydrolysis of afzelin and quercitrin, respectively [19 (link)]. Kaempferol-7-O-glucoside, quercetin-7-O-glucoside, rutin (quercetin-3-O-rutinoside), isorhamnetin, and isorhamnetin-3-O-glucoside were purchased from Yuanye Bio-Technology (Shanghai, China). Potassium peroxodisulfate (K2S2O8), phosphoric acid (H3PO4), sodium phosphate monobasic dihydrate (NaH2PO4·2H2O), disodium hydrogen phosphate dodecahydrate (Na2HPO4·12H2O), ABTS, and DPPH were purchased from J&K Scientific (Beijing, China). Ultra-pure water was prepared using a Millipore Milli-Q system (Darmstadt, Germany). All water used was ultrapure unless otherwise noted. Methanol, acetone, acetonitrile, and formic acid (LC-MS grade) were purchased from Fisher Scientific (Pittsburgh, PA, USA). All solvents were degassed with dry nitrogen to remove dissolved O2 and CO2 before use.

Phytochemical Analysis and Anticancer Activity

The fresh PPL was collected from Hangzhou City (Zhejiang, China) in 01 September, 2020. These samples were identified by Professor Wang Ping from Zhejiang University of Technology. The voucher specimens (No. PPL 20200923) were deposited in Moganshan Campus of Zhejiang University of Technology. The leaves were air−dried naturally at room temperature, crushed, and passed through an 80−mesh sieve. After that, they were stored in a −18 °C freezer until use.
The standards of polyphyllin I (≥98%), II (≥98%), VI (≥98%), and VII (≥95%); dioscin (≥98%); kaempferol 3-O-gentiobioside (≥95%); isorhamnetin-3-O-glucoside (≥98%); and cisplatin were purchased from Shanghai Yuanye Biotechnology Co. Ltd., (Shanghai, China). Ultrapure water was produced by a Barnstead TII Pure Water System (Waltham, MA, USA). All other analytical−grade chemicals used in this experiment were purchased from Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China). A549 cells, MCF−7 cells, HepG2 cells, A431 cells, and HBE cells were purchased from Hunan Fenghui Biotechnology Co. Ltd. Roswell Park Memorial Institute (RPMI−1640). Dulbecco’s Modified Eagle Medium (DMEM) and fetal bovine serum (FBS) were obtained from Gbico (New York, NY, USA).

(Shanghai, China). Mearnsetin and laricitrin were from ChemFaces (Wuhan, China). Methyl 3-O-methylgallate was from MedChemExpress (Monmouth Junction, NJ, USA). Myricetin 3′, 4′-dimethyl ether was provided by Prof. Ai-Xia Chen of Shandong University, China.
