Ain93m

Manufactured by Bio-Serv

The AIN93M is a laboratory animal diet formulated to meet the nutritional requirements of laboratory rodents, such as mice and rats, during the maintenance phase of their life cycle. It provides a balanced source of proteins, carbohydrates, fats, vitamins, and minerals to support the general health and well-being of laboratory animals.

BALB/cJ (#00651), C57BL/6J (#00664), Agouti lethal yellow
(Ay/a; C57BL/6J;#002468) and UCP1−/−C57BL/6J;#003124) mice were obtained from Jackson Labs and bred in-house. All
animals were housed at constant temperature (19 −23°C) in 12 h
light/dark cycles with free access to water and standard chow (Harlan Teklad
Global Diet 2020X). For high fat feeding, C57BL/6J mice were provided with
Harlan Teklad diet#TD06415 (45% calories from fat); for calorie restriction
(CR), ad libitum consumption rates of chow
were measured for singly housed mice; then mice were 25% calorie restricted
using a 60% mix of AIN93M:40% CR (Bioserv cat#F05314) in control diet (AIN93M,
Bioserv cat#F05312), pair-fed daily at 5 pm, for 5 weeks.

Synthesis of V-125 was previously described31 (link). Bexarotene was purchased from LC Laboratories. Rexinoids were dissolved in a vehicle (50 ml/kg diet) of 1 part ethanol:3 parts highly purified coconut oil (Neobee oil, Thermo Fisher) and then mixed into 1 kg of powdered diet (AIN-93M, BioServ for A/J mice or 5002 rodent chow, PMI Nutrition International for MMTV-neu mice) using a commercial KitchenAid mixer. The same vehicle was used in the control diets. In the MMTV-Neu model, doses were 30 mg per kg of diet (~ 7.5 mg per kg body weight) for prevention studies and 100 mg per kg of diet (~ 25 mg per kg body weight) for treatment studies, as utilized in previous publications20 (link),32 (link). In the A/J model, doses were 40 and 80 mg per kg of diet (~ 10 and 20 mg per kg body weight), doses used in previous publications25 (link). Rexinoids remain stable in diet at 4 °C for up to 6 weeks, as confirmed by liquid chromatography-mass spectrometry.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
