Cell Culture Protocol for MDCK and HEK293T

Human renal cell line HEK293T cells (ATCC® CRL-3216TM) were grown in Dulbecco’s modified Eagle medium (DMEM; 12100-046, Gibco, United States) supplemented with 10% FBS and antibiotics as described above.

Comparative Efficacy of Influenza and RSV Growth

Madin-Darby Canine Kidney (MDCK) cells (ATCC number, CCL-34TM) and HEp-2 cells (ATCC number, CCL-23TM) were purchased from ATCC (Manassas, VA 20110, USA.) and used for the propagation and initial detection of viruses based on cytopathic effect. Eagle Minimal Essential Medium (EMEM) (Gibco, USA) supplemented with 10% Fetal Bovine Serum (FBS) (Gibco, USA) were used to grow the MDCK and HEp-2 cell lines. These were used in combination to observe the comparative efficacy for the growth of both influenza and RSV. Method of Ken et al.11 was followed to set up of assay.

MDCK Cells Influenza Virus Assay

The Madin–Darby Canine Kidney (MDCK) cells were obtained from ATCC® CCL-34TM. H1N1 influenza virus was provided by the Virus Laboratory, Guangzhou Women and Children’s Medical Center, Guangzhou Medical University. Fetal bovine serum and DMEM were purchased from Thermo Fisher Scientific, Waltham, MA, USA. Na2SeO3, vitamin C, AM, MTT, propidium iodide, 4′,6-diamidino-2-phenylindole (DAPI), and 2′,7′-dichlorofluorescin (DCF)-diacetate were purchased from Sigma-Aldrich Co. (St Louis, MO, USA). AKT and caspase-3 antibodies were purchased from Cell Signaling Technology (Boston, MA, USA).
