Multi-Photon Imaging of Collagen Fibers

Check if the same lab product or an alternative is used in the 5 most similar protocols
A TriM Scope II multi photon system from LaVision BioTec was used for SHG imaging to visualise anisotropic structures, such as collagen fibres. The microscope setup is a single beam instrument with an upright Olympus BX51 WI microscope stand equipped with high sensitive non-descanned detectors close to the objective lens. The TriM Scope II is fitted with a Coherent Scientific Chameleon Ultra II Ti:Sapphire laser (tuning range 680-1080 nm) and a Coherent Chameleon Compact OPO (automated wavelength extension from 1000 nm to 1600 nm). A 20x IR objective lens (Olympus XLUMPlanFl 20x/1.0W) with a working distance of 2.0 mm was used. SHG signals were detected using TiSa light at 850 nm, a 420/40 band pass filter and a blue-sensitive photomultiplier (Hamamatsu H67080-01). 3D-images were acquired and processed with LaVision BioTec ImSpector Software under Microsoft Windows. Overview images were reconstructed from a sequence of tiled 3D images with Image J.

Visualizing Bone Microstructure with Multiphoton Microscopy

Check if the same lab product or an alternative is used in the 5 most similar protocols
We used a TriM Scope II multi photon system from LaVision BioTec to visualize immune labeling and SHG generated by collagen in the bone. The setup is a single-beam instrument with an upright Olympus BX51 WI microscope stand that is equipped with high sensitive non-descanned detectors close to the objective lens. The TriM Scope II is fitted with a Coherent Scientific Chameleon Ultra II Ti:Sapphire laser and a Coherent Chameleon Compact optical parametric oscillator (OPO). A 20× infrared (IR) objective lens (Olympus XLUMPlanFL; numerical aperture [NA] 1.0) with a working distance of 2.0 mm was used. The microscope is equipped with a pair of x-y galvanometric mirrors used to scan at a scanning speed of up to 1,200 lines/s. The maximal laser power (850 nm) on the object was 10–20 mW at superficial areas and 80–100 mW when imaging at large depths. Dichromatic mirrors and band pass filters spectrally separate the emitted light before the signal is detected using attached photomultiplier tubes (PMTs; Hamamatsu H67080-01 (blue channel), and H67080-20 (green and red channels). The following detection channels were used: red (560–680 nm), green (475–575 nm), and blue (370–470 nm). 3D images were acquired and processed with LaVision BioTec ImSpector Pro.

Visualizing Skeletal Muscle Fiber Organization

Check if the same lab product or an alternative is used in the 5 most similar protocols
A TriM Scope II multi photon system from LaVision BioTec was used to visualize skeletal muscle fiber organization inside organoids and distinct sarcomeres. The microscope setup is a single beam instrument with an upright Olympus BX51 WI microscope stand equipped with highly sensitive nondescanned detectors close to the objective lens.
Muscle fiber SHG signals were detected in forward direction using TiSa light at 850 nm, a 420/40 band pass filter and a blue-sensitive photomultiplier (Hamamatsu H67080-01). 3D-images were acquired and processed with LaVision BioTec ImSpector Software.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
