Pseudoternary Nanoemulsion Formulation of ASPS

The ASPS-NE formulations were synthesized using the method of pseudoternary phase diagram (Yu et al., 2020 (link); Zhao et al., 2020 (link)). On the basis of the nanoemulsion regions in the phase diagram, ethanol was selected as the co-surfactant (Germany BASF Co., Ltd). Tween-80 and span-80 (Damao Chemical reagent factory, Shanghai, China) were selected as surfactants (Cao et al., 2019 (link); Wang et al., 2018 (link)). Liquid paraffin (Damao Chemical reagent factory, Tianjin, China), Isopropyl myristate (IPM, Lanxi Wumart Chemical Co., Ltd., Zhejiang, China) was selected as the oil phase, Acanthopanax senticosus polysaccharide (ASPS: purity =98%, average MW =30.51 kDa) was provided by Shanghai Yuanye Biotechnology Co., Ltd. First, k-Car-ASPS was dissolved in deionized water, and the resulting aqueous solution was then mixed with ethanol and liquid paraffin, followed by the addition of respective amounts of Tween-80, Span 80, and IPM. The clear and transparent nanoemulsion was obtained after magnetic stirring. The final NE formulation comprised 46.2% Tween-80, 15.4% Span 80, 7.6% ethanol, 15.4% liquid paraffin, 11.5% IPM, and 3.9% aqueous ASPS solution.

Characterization of E. ulmoides Seed Oils

E. ulmoides seed oils were purchased from Jiangxi Jingwang Natural Fragrance Co., Ltd., Xi'an Huageng Biotechnology Co., Ltd., Shaanxi Nexant Biotechnology Co., Ltd., and Jiangxi Hai Lin Fragrance Co., Ltd. Chitosan (CS) and gum arabic (GA) were acquired from Shanghai Macleans Biochemical Company. Hydroxyethyl cellulose (HEC) was obtained from Ashland Company. Span-80, maltodextrin (MD), sodium alginate (SA), soy protein isolate (SPI), and gelatin (Ge) were purchased from Shanghai Yuanye Biotechnology Co., Ltd. Transglutaminase was obtained from Taixing Dongsheng Food Co., Ltd.
Caprylic/capric triglyceride was purchased from Croda Cypro Chemical (Sichuan) Co., Ltd. Hydrochloric acid and sodium hydroxide (analytical grade) were purchased from Xilong Chemical Co., Ltd. Methanol (pure for mass spectrometry) was acquired from Fisher Scientific World, USA. N-hexane (analytically pure) was obtained from Shanghai Aladdin Biochemical Technology Co., Ltd. Finally, a 14% boron trifluoride methanol solution was purchased from Fuchen Chemical Reagent Co., Ltd.

Chitosan-Based Antioxidant Biomaterials

Dulbecco's Modified Eagle Medium (DMEM), Phosphate Buffer Saline (PBS, 1×), fetal bovine serum (FBS), and trypsin–EDTA (0.25% trypsin and 0.913 mM EDTA tetrasodium) were purchased from Gibco (USA). Mouse fibroblast cells (L929) were purchased from the Cell Bank of the Chinese Academy of Sciences (Shanghai, China). Cell Counting Kit-8 (CCK-8) was brought from Beyotime Biotechnology. A live & dead viability/cytotoxicity assay kit was obtained from YEASEN Inc (Shanghai, China). Sprague Dawley (SD) rats and balb/c mice were supplied by the affiliated hospital of Qingdao university laboratory animal center. Ultrapure water was supplied by Milli-Q Plus System (Millipore Corporation, United States). All reagents were used without further purification.
