Engineered E. coli Phenylpropanoid Production

Eriodictyol, dihydrokaempferol (DHK), and DHQ were purchased from Merck (Sigma Aldrich). p-Coumaric acid and naringenin were purchased from Tokyo Chemical Industry. TT7 (codon optimized for E. coli) was synthesized as gBlocks Gene Fragment from Integrated DNA Technologies Inc. The ATR2 gene (codon optimized for E. coli) was synthesized from GenScript. All strains and plasmids used in this study are listed in Table 1 and2, respectively. E. coli DH5α (Invitrogen) was used for routine gene cloning works. BTY5 (Kim et al., 2018) was used as a host strain for the production of all phenylpropanoids.

Folin-Ciocalteu reagent, sodium carbonate, and p-hydroxyacetophenone were purchased from Nacalai Tesque (Kyoto, Japan) . Fluorescein sodium salt and 1,1-diphenyl-2-picrylhydrazyl (DPPH) were purchased from Sigma-Aldrich (St. Louis, MO., U.S.A.) . p-Coumaric acid was purchased from Tokyo Chemical Industries (Tokyo, Japan) . Schaftoside and isoschaftoside were isolated from sugarcane products and identified based on NMR and UV-Vis spectra previously 8) . All other reagents were from Wako Pure Chemical Industries.
