Preparation of Agonist Stock Solutions

Check if the same lab product or an alternative is used in the 5 most similar protocols
Ado and ACh (Sigma-Aldrich, St Louis, MO, USA) were dissolved in deionized water to make 10 mM stock solutions and stored at -20°C. Working agonist concentrations were freshly prepared in the recording extracellular solution. E-4031 and chromanol 293B were obtained from Tocris Bioscience (Ellisville, MO, USA), K4BAPTA was acquired from Santa Cruz Technology (Dallas, TX, USA), and all other reagents were from Sigma-Aldrich.

Preparation of PIP2 and SPM solutions

Check if the same lab product or an alternative is used in the 5 most similar protocols
SPM was obtained from Sigma-Aldrich, dissolved in water as a 100-mM stock solution, and diluted in the bath solution just before use. Brain phosphatidylinositol 4,5-bisphosphate (PIP2; dissolved in chloroform:methanol:water) was purchased from Avanti Polar Lipids, aliquoted, dried under a nitrogen stream, and stored at −80°C. Before the experiment, PIP2 was dissolved in FVPP solution by sonication for 10 min to obtain a concentration of 10 µM. All other reagents were purchased from Sigma-Aldrich, except K4BAPTA (Santa Cruz Biotechnology).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
