Gentamycin sulfate

Human ovarian adenocarcinoma cell (HOAC) lines NIH-OVCAR-3 and SKOV-3 (ATCC® numbers HTB-161 and HTB-77) were obtained from the American Type Culture Collection (Manassas, VA). HOAC line IGROV-1 was a gift from the Gustave Roussy Institute, Villejuif. A2780 cells were obtained from Sigma Aldrich (France). EFO-21 cell line was obtained from the Deutsche Sammlung von Mikroorganismen und Zellkulturen GmbH (DSMZ, Germany). HOAC were cultured in RPMI 1640 medium containing 10% of fetal bovine serum, supplemented with with 10% FBS, 10 mM HEPES, 2 mM L-glutamine, gentamycin sulfate and fungizone (all reagents purchased from Wisent Inc., St-Bruno, Canada). Cell lines were maintained as monolayers at 37 °C in a humidified 5% CO₂ atmosphere.

HGSOC cell lines were cultured in 100 mm petri dishes (Sarstedt Inc., Nümbrecht, Germany) in OSE medium (WISENT Inc., St-Bruno, QC, Canada) supplemented with 10% fetal bovine serum (WISENT Inc.), 0.5 µg/mL amphotericin B (WISENT Inc.) and 50 µg/mL gentamycin sulfate (WISENT Inc.) (complete OSE medium). Plates were maintained at 37 °C in low oxygen conditions (7% O₂ and 5% CO₂). Cells were passaged at near confluence by trypsin 0.05% (WISENT Inc.) digestion. Cultures were discarded before the 20th passage, after which a fresh batch of cells was thawed for further experiments. For resistant cell lines, passages were counted from when stable resistance was confirmed.

Jurkat T lymphoma cells were kindly provided by Dr. Lapointe Réjean (CRCHUM), while MCF-7, SKOV3, and all PC cell lines (22Rv1, LNCaP, DU145, and PC3) were obtained from the American Type Culture Collection (ATCC, Manassas, VA, USA). Jurkat T lymphoma cell and PC cells were maintained in RPMI 1640 medium (Wisent Inc., St-Bruno, QC, Canada), MCF-7 was grown in DMEM medium (Wisent Inc.), and SKOV3 in OSE medium (Wisent Inc.). All culture media were supplemented with 10% fetal bovine serum (FBS) (Gibco^®, Thermo Fisher Scientific, Waltham, MA, USA), 0.454 μg/mL of amphotericin B (Wisent Inc.), and 90 μg/mL gentamycin sulfate (Wisent Inc.).
