Cultivation of Murine Melanoma and Stem Cells

The murine melanoma cells B16-F10 were purchased from ATCC and cultured in DMEM/HG (HyClone, USA) supplemented with 10% FBS (Gibco, USA). Human induced pluripotent stem cells (DYR0100) were purchased from ATCC and cultured in mTeSR1 medium (STEMCELL Technologies, Canada). IL-24-iPSCs was previously generated by our group. The MSCs derived from iPSCs were cultured in MSC medium with DMEM/LG (HyClone, USA) supplemented with 10% FBS and 0.1% bFGF (Sigma, USA). All cells were cultured at 37 °C in a humidified chamber maintained at 5% CO2.

Reprogramming and Culturing of hiPSCs and iMSCs

The normal hiPSCs (DYR0100) were purchased from ATCC. HA-iPSCs [32 (link)] and T-7-iPSCs [33 (link)] were generated previously. Briefly, HA-iPSCs were reprogrammed by urine cells from a severe HA patient with F8 intron 22 inversion. T-7-iPSCs were obtained by site-specific integration of CMV-driven BDDF8 at the rDNA locus in HA-iPSCs. All hiPSCs were routinely cultured (37 °C, 5% CO2) on Matrigel- (BD Biosciences, Franklin Lakes, NJ, USA) coated 12-well plates (Corning, New York, NY, USA) in mTesR Plus medium (StemCell Technologies, Vancouver, BC, Canada).
The iMSCs were routinely cultured (37 °C, 5% CO2) on 0.1% gelatin (StemCell Technologies, Vancouver, BC, Canada) coated 10-cm dishes (Corning, New York, NY, USA) in αMEM basal medium (Thermo Fisher Scientific, Waltham, MA, USA) containing 10% FBS (Gibco, New York, NY, USA), 2 mM GlutaMAXTM (Gibco, New York, NY, USA), and 0.1% bFGF (Gibco, New York, NY, USA).

Generation and Culture of Induced Pluripotent Stem Cells

The normal hiPSCs (DYR0100) were purchased from ATCC. DMD-iPSCs were generated previously [20 (link)]. Briefly, DMD-iPSCs were reprogrammed from urine cells of a severe DMD patient with exon 50 deletion. All iPSCs were routinely cultured (37 °C 5% CO2) on Matrigel (BD Biosciences, Franklin Lakes, NJ, USA) coated 12-well plates (Corning, New York, NY, USA) in mTesR1 medium (StemCell Technologies, Vancouver, BC, Canada).
The HEK 293T cells were routinely cultured (37 °C, 5% CO2) on 6-well plates (Corning) in DMEM basic medium (Gibco, New York, NY, USA) containing 10% FBS (Gibco), 2 mM GlutaMAXTM (Gibco).
