Calbindin 28kDa Immunofluorescence Staining

Following fixation, cultures were permeabilized in 0.1M PBS containing 0.02% Triton X-100 (SIGMA) for 30 minutes at room temperature and subsequently blocked for 1 hour with 10% goat or horse serum (Vector Laboratories). Tissue was incubated in primary antibody overnight at 4°C at the listed dilutions. Antibody staining was visualized using secondary antibodies conjugated to either Alexa488 orAlexa 546 (Invitrogen). Cultures were incubated with all secondary antibodies for 1 hour at room temperature, washed in PBS and then incubated for an additional hour with either Alexa Fluor-conjugated Phalloidin-546 or-633 (Invitrogen) to label filamentous actin.
