Formulation and Characterization of Peptide-Loaded Nanoparticles

Phytantriol was purchased from A & E Connock (Hampshire, England). Poloxamer 407 (Lutrol® F127) was obtained from BASF (Ludwigshafen, Germany) and 1,2-propandiol (Propylene Glycol, purity≥99.5%) from Merck (Darmstadt, Germany). CD8 Ovalbumin peptide (SIINFEKL, OVA257–264) and CD4 Ovalbumin peptide (ISQAVHAAHAEINEAGR, OVA323–339) and 5(6)-tetramethylcarboxy rhodamine-labelled OVA257–264 (TMR-SIINFEKL) was purchased from Mimotopes (Clayton, Australia). Monophosphoryl Lipid A from Salmonella Minnesota RE 595 was purchased from Sigma-Aldrich (Missouri, USA). Purified saponin (Quil-A) was purchased from Brenntag Biosector (Frederikssund, Denmark). All other chemicals were of analytical grade.

Artemether Topical Formulation Development

Artemether (ART) was procured from Ipca Laboratories Ltd., India. Gelucire ® 43/01 Pellets and Transcutol ® P were obtained from Gattefossé, France whereas Phospholipon ® 85G (P85G) was donated by Lipoid GmbH, Germany. Tween ® 80, sodium azide and sorbitol were obtained from Sigma-Aldrich, USA. Formulation and Characterization of β-Lapachone

β-Lapachone was supplied by the Mazence Pharmaceutical Company (Suwon, Korea). Crospovidone (Kollidon CL), poloxamer 407 (Lutrol F 127), and poloxamer 188 (Lutrol F 68) were obtained from BASF (Ludwigshafen, Germany). Poly(acrylic acid) (Carbopol 940) was procured from Lubrizol Advanced Materials Inc. (Cleveland, OH, USA). Xanthan gum, hydroxypropyl methylcellulose (HPMC), sucrose, and poly(ethylene oxide) (PEO) were purchased from the Sigma-Aldrich Company (St. Louis, MO, USA). Polyethylene glycol (PEG) 4000 was obtained from Daejung Chemicals and Metal Co., Ltd. (Siheung, Korea). Polydextrose was purchased from Santa Cruz Biotechnology Inc. (Dallas, TX, USA). Spray dried lactose was kindly provided by Whanin Pharm Co., Ltd. (Suwon, Korea). All other materials used were of pharmaceutical grade.

Poloxamer 407-Based Nanocarrier Extraction

Poloxamer 407 (Lutrol F127) was from BASF AG (D-Ludwigshafen), cholesterol, sucrose, tween 80 and MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium Bromide) were from Sigma-Aldrich (D-Steinheim), egg phosphatidyl choline (EPC) was obtained from Lipoid GmbH (D-Ludwigshafen), monoolein (GMOrphic-801) from Eastman Chemical Company (Kingsport, TN), methanol was from Carl Roth GmbH (D-Karlsruhe), ethanol and chloroform all from VWR International (D-Darmstadt) , and Hepes and sodium chloride were from AppliChem GmbH (D-Darmstadt). The cell lines were purchased from ATCC, USA. All cell culture materials were purchased from Lonza Bioscience (Morristown, USA). Puri ed water was prepared by ltration and deionization/reverse osmosis (Milli RX 20, Millipore, D-Schwalbach) Plant collection Ficus carica L. fam. Moraceae was collected from Wadi Fatima, Makkah, KSA and identi ed by Dr. Hany Gouda, Department of Pharmacognosy, Faculty of Pharmacy, Najran University. A voucher specimen (UQU-2019-1) of the plant is available in the herbarium, Department of Pharmacognosy, Faculty of Pharmacy, Umm al-Qura University. Dried plant leaves were powdered and extracted with petroleum ether. The obtained extract was evaporated and then subjected to separation using preparative HSCCC.
