Plasma-deposited Antibacterial Coatings

Glass plates (soda-lime glass, 150 × 100 mm, thickness 1.1 mm) were used as substrates for deposition. 2-Methyl-2-oxazoline (98%, Sigma-Aldrich, Munich, Germany) was used as a monomer for plasma deposition. Preliminary antibacterial tests were done with Staphyloccocus epidermidis (CCM 4418), supplied by the Czech Collection of Microorganisms in Brno. Bacterial culture was grown into commercial BHI medium (Brain Heart Infusion Broth, HiMedia, Mumbai, India). Basic Red 2 (Safranin O, Sigma-Aldrich, Munich, Germany) stain was used for biofilms visualization. Other antibacterial tests were done with Staphylococcus aureus (CCM 4516) and Escherichia coli (CCM 4517), both supplied by the Czech Collection of Microorganisms in Brno.

Synthesis of Polyoxazolines

2-methyl-2-oxazoline (MeOx), 2-ethyl-2-oxazoline (EtOx), methyl p-tosylate (MTS), calcium hydride (CaH2), and acetonitrile (AN) were received from Sigma-Aldrich (St. Louis, MO, USA). 2-isopropyl-2-oxazoline (IPOx) was received from Tokyo Chemical Industry co (Fakuya, Japan). Potassium hydroxide (KOH) and methanol were obtained from LachNer (Neratovice, Czech Republic). MeOx, EtOx, IPOx, and AN were distilled over CaH2. MTS was used as received.

Synthesis and Characterization of Functionalized Nanoparticles

2-Methyl-2-oxazoline, NaOH pellets, phosphate buffer saline (PBS) tablets, foetal bovine serum (FBS), 2-mercaptosuccinic acid (MSA) (97%), sodium borohydride (NaBH4), nitric acid (70%) were purchased from Sigma-Aldrich Australia and used as received. Hydrochloric acid (36%, Ajax Finechem Pty. Ltd. Australia) was used as received. Dulbecco's Modified Eagle Medium (DMEM) was purchased from Gibco. Penicillin, streptomycin, l-glutamine, MEM non-essential amino acids, cell counting kit-8 (CCK-8), Naphthol AS-MX phosphate and Fast Red TR were obtained from Sigma. Silver nitrate (AgNO3) and glass coverslips were bought from ProSciTech, Australia. Ultra-pure MilliQ water (resistivity 18.2 Ω) was used for all experiments and cleaning procedures.

Synthesis and Characterization of Oxazoline-Based Polymers

The following materials were purchased from Thermo/Fisher: WhatMan Anotop 0.025 μm non-sterile filters, 25 gauge 0.5 inch, long blunt needle (Strategic Applications Inc., Lake Villa, IL, USA), 40% acrylamide 29:1, TEMED, Ammonium persulphate, Malvern Folded Cap Cells, DLS cuvettes (Malvern Pananalytical, Malvern, UK), radioimmunoprecipitation assay buffer, HALT protease and phosphatase inhibitor cocktail, SuperSignal™ West Pico PLUS Chemiluminescent Substrate, iBRIGHT prestained protein ladder, Geneticin™ Selective Antibiotic (G418 Sulfate), Neurobasal Media, B27 supplement, Glutamax 100× acetonitrile, potassium carbonate, diethyl ether, methanol, agarose tablets, and sodium hydroxide. BDNF was obtained from Novoprotein, which later became Bon Opus Biosciences. PSR-PLE was obtained from Peptide Solutions LLC. Colorado calf serum was obtained from the Colorado Serum Company. We obtained Bio-Safe Coomassie Stain and Native Sample buffer for protein gels 4× from Bio-Rad, Hercules, CA, USA.

Plasma Polymerization of Oxazoline Coatings

Deposition of plasma polymers was carried out in a custom-made reactor that has been previously described previously (Chan K. M. et al., 2020 (link)). The plasma was ignited with a 13.56 MHz Radio Frequency generator coupled to a matching network (Coaxial Power System). Substrates, including glass microscope slides, silicon wafers and coverslips, were introduced into the reactor and primed with an air plasma (30 W, 3 min, 0.12 mbar). The reactor was evacuated to a base pressure of 2 × 10−2 mbar to remove atmospheric oxygen and nitrogen from the chamber. The pressure was monitored with a pirani gauge (APG100, Edwards) and controlled with a needle valve (Chell Instrument, United Kingdom). The precursors, 2-methyl-2-oxazoline and 2-isopropenyl-2-oxazoline (Sigma Aldrich, Australia) were introduced into the reactor at a working pressure of 0.12 mbar at which point the plasma was ignited at 30 W. Deposition was conducted for 30 and 50 s after which the monomer flow was stopped using the needle valve.
For integration into the spiral microfluidic, PPOx coatings were deposited on glass microscope slides (Marienfeld, Germany) through a custom-made aluminum mask to limit the film deposition to the cell capture reservoir area.

Cell media and all other chemicals were purchased from Sigma (Buchs, Switzerland): N-(2-hydroxyethyl)phthalimide (99%), trifluoromethanesulfonic anhydride (≥ 99%), triethylamine (≥ 99%), 2-methyl-2-oxazoline (98%), sodium azide (≥ 99.5%), hydrazine monohydrate (98%), β-benzyl-L-aspartate (98%), triphosgene (98%), DET (99%), calcium hydride (95%), acetonitrile (≥ 99.8%), dichloromethane (≥ 99.8%), ethanol (99.8%), N,N-dimethylformamide (≥ 99.8%), and tetrahydrofuran (≥ 99.9%). triethylamine, dichloromethane, 2-methyl-2-oxazoline, and acetonitrile were separated from CaH 2 , and tetrahydrofuran was separated from LiAlH 4 by distillation. Other chemicals and solvents were used without additional purification.
