CLN from PBS-perfused mice were snap-frozen in Tissue-Tek OCT compound (Sakura Finetex, Torrance, CA) and sectioned at 10 μm using a Thermo Shandon cryostat. Sections were fixed with ice cold 70% acetone for 5 min, blocked with 5% bovine serum albumin and 10% goat serum for 1 h and then stained with rat anti-mouse FITC conjugated GL7 mAb (BD Biosciences), rat anti-mouse B220 mAb (BD biosciences) and rabbit anti-mouse CD3 polyclonal Ab (Abcam, Cambridge, MA) overnight at 4 °C.
