Transcriptome analysis of plant leaves

RNA extraction and cDNA library preparation and high throughput sequencing Whole plants were harvested in RNAlater TM (ThermoFisher Scientific). Once all samples were collected, leaf 3 primordias and leaf 3 blades were dissected with a scalpel, in a Petri dish filled with RNA later solution, under a Leica MZ 16 F dissecting microscope. Total RNA was extracted using the RNeasy Plant Mini Kit (Qiagen) with on-column DNase digestion. Samples were then sent to Edinburgh Genomics for QC check and sequencing.
