Glucose and Lactate Assays in HSC6 and OECM1 Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Glucose Uptake Assay Kit (KA4086) and Lactate Assay Kit (KA0833) were obtained from Abnova (Taiwan, China). Basing on the kit instructions, the glucose uptake and lactate production of HSC6 and OECM1 cells were determined.

Monocyte chemotactic protein (MCP-1), tumor necrosis factor α (TNFα), and interleukin-6 (IL-6) levels were analyzed by sandwich ELISA (BioCheck, Foster City, CA). In addition, a glutathione peroxidase (GPx) assay kit, glutathione (GSH) assay kit, and superoxide dismutase (SOD) assay kit were used to evaluate GPx, GSH, and SOD levels, respectively. Caspase 3 activity was monitored using a caspase 3 assay kit (Beyotime Institute of Biotechnology). A glucose uptake assay kit was used to measure glucose uptake (Abnova, Taiwan, China; KA4086), while a lactate assay kit was used to determine concentrations of lactate (Abcam, Cambridge, UK; ab65331).

Intracellular ATP production was measured using an ATP Assay Kit (Abcam, United States). Lactate production were measured using L-Lactate Assay Kit (Abcam, United States) and glucose uptake were measured using a Glucose Uptake Assay Kit (Abnova, China) according to the manufacturers’ instructions, respectively.

Glucose Uptake Quantification in LX2 Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
The glucose uptake by LX2 cells was determined using a Glucose Uptake Assay Kit (Abnova, Taiwan, China) according to the manufacturer’s instructions. In this assay, the glucose analog 2-DG is metabolized to 2-DG-6-phosphate, which is proportional to glucose uptake by cells. The accumulated 2-DG-6-phosphate is enzymatically coupled to generate NADPH, which is specifically monitored by a NADPH sensor. The signal can be read by an absorbance microplate reader by reading the OD ratio at wavelength 570 to 610 nm.

Glucose Uptake Modulation by Anx A5 and Cytokines

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cells were seeded into a 96-well black wall bottom cell culture plate and treated with various concentrations of Anx A5 and 10 ng mL−1 LPS&IFN-γ or 20 ng mL−1 IL-4 for 6 h prior to measuring glucose uptake. Cells were grown in glucose-free DMEM medium (Gibco BRL, #11966–025) for additional 1 h. Glucose uptake was measured using glucose uptake assay kit (Abnova, KA4086) according to the manufacturer's instruction.

Glucose Uptake Assay with P7C3

Check if the same lab product or an alternative is used in the 5 most similar protocols
Glucose Uptake Assay Kit (#KA4086, Abnova, Taiwan) was used for glucose uptake measuring. First, 5 × 104 cells/well/100 μL/were seeded in a 96-well plate in treatment medium which containing 0.3% DMSO (0 μM P7C3), 30 μM P7C3, or 50 μM P7C3 for 18 h. The medium was then replaced with a new growth medium, and the cells were cultured for another 6 hours before experiments. Then, the cells were treated as desired, and 10 μL/well 2-Deoxy-d-glucose was added, and the cells were incubated at 37°C for 30 min. To wash and lyse cells, 50 μL/well 2-Deoxy-d-glucose uptake assay working solution was added, and the cells were incubated at RT for 90 min. The optical density (OD) ratio at 570/610 nm was measured using a spectrophotometer (1510, Thermo Fisher, USA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
