Single-cell suspensions of BMDMs or BMDCs were blocked with FcBlock (anti-CD16/CD32; BioLegend) and subsequently stained with fluorescence-labeled mAbs for CD11b, CD11c, Gr-1, Ly6C, TLR4/MD2 (clone MTS510), and TLR4 (clone SA15-21; all BioLegend). Intracellular staining of human and mouse TNF (both BioLegend) and mouse TLR3 (Imgenex) was performed in permeabilized cells according to standard procedures. Flow cytometric measurements were performed on a FACSCantoII cell analyzer and an LSRII flow cytometer (both BD). Data were analyzed with FlowJo software (version 9.8.3, Mac; version 10.0.7, PC; Tree Star).

Immune Cell Profiling in Mouse Peritoneum

Peritoneal cavity of mice was flushed with 8 ml of PBS. Cells were pelleted, washed, and re-suspended in FACS buffer (PBS, 5 mM EDTA, 2.5% FBS) for immunostaining and flow cytometric analyses. Titrated amounts of the following antibodies from BioLegend were used: CD45 (clone 30-F11), CD11b (clone M1/70), F4/80 (clone BM8), CD19 (clone 6D5), CD5 (clone 53–7.3) and TLR4 (clone SA15–21). Data were acquired using FACSVerse (BD) and analyzed using FlowJo software.
