Measuring Putrescine Uptake in T Cells

T cells (2 × 106) were suspended in 200 μl of PBS containing permeant putrescine (100 μM) and [1,4-14C]-putrescine dihydrochloride (0.2 μCi, ARC 0245) and then incubated at 37°C for 10 min (within the established linear phase of uptake). The uptake was stopped by loading all the transport mixture onto a discontinuous gradient of bromododecane and perchloric acid/sucrose and then centrifuged at 14, 000g for 90 s. The discontinuous gradient was prepared by overlaying 1-bromododecane (800 μl, Sigma-Aldrich) above 20% perchloric acid (100 μl, Sigma-Aldrich)/8% sucrose solution in a 1.5-ml microfuge tube. The samples were snap frozen in an ethanol-dry ice bath. The bottom part of the microfuge tubes containing T cell lysate in perchloric acid–sucrose was cut by a microfuge cutter. The cell lysates were diluted with 300 μl of 0.5% SDS–1% Triton X-100, and then transferred into scintillation vials with 10 ml of scintillation cocktail. The radioactivity was quantitated by liquid scintillation spectrometry.

Synthesis of Alkylated Gold Nanoparticles

from Sigma-Aldrich. Potassium hexachloroiridate(IV) (39.8% Ir) was
obtained from Alfa Aesar. Sodium thiosulfate pentahydrate (Na2S2O3·5H2O) along with
solvents such as toluene, acetone, ethanol, and methanol was purchased
from Fisher Scientific. Chloroform-d was obtained
from Cambridge Isotope Laboratories. Sodium S-dodecylthiosulfate
was synthesized using the published procedure.11 ,18 Water was purified by using a Barnstead Nanopure Diamond ion exchange
resins purification unit.

Synthesis of Imidazolium Ionic Liquid Surfactants

from Sigma-Aldrich Co. The reagents used such as isopropanol and ethyl
acetate were purchased from Tianjin Kemiou Reagent Co. In all experiments,
deionized water was used to configure the solution. Crude oil was
obtained from one of CQ oil reservoirs. The viscosity of crude oil
was 1.5 mPa·s at a reservoir temperature of 65 °C.

Trypsin, fetal bovine serum (FBS), Dulbecco’s Modified Eagle’s Medium (DMEM), and Roswell Park Memorial Institute (RPMI) 1640 medium were all provided by GE Healthcare UK Ltd (Little Chalfont, UK). 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) was from AMRESCO LLC (Solon, OH, USA). LysoTracker® Red dye, Hoechst 33258, apoptosis detection kit (C1062), and Bradford protein assay kit (P0006) were all from Beyotime Institute of Biotechnology (Shanghai, People’s Republic of China). Luciferase assay kit (E1500) was from Promega Corporation (Fitchburg, WI, USA). Other chemicals and reagents were of analytical grade. Mouse mammary tumor cell line 4T1 was provided by the Department of Pathology in Institute of Medicinal Biotechnology in Peking Union Medical College (Beijing, People’s Republic of China) and cultured in RPMI 1640 containing 10% FBS.

Purification and Characterization of Organic Compounds

and potassium hydroxide (85%) were delivered by Avantor (Gliwice,
Poland) and used without further purification. Deionized water with
a conductivity < 0.1 μS cm–1, from the
HLP Smart 1000 demineralizer (Hydrolab, Poland), was used.

Synthesis of Dodecyl Derivatives

Synthesis of Polyketones and Novel Compounds
