Kinetic Titration of Aβ(1-42) with scFv IC16

Quantifying USP4 DUSP-Ubl Binding

Concentration series of USP4 DUSP–Ubl in running buffer (20 mM HEPES pH 7.5, 150 mM NaCl, 5 mM DTT and 0.05% Tween 20) supplemented with NSB reducer (GE Healthcare) were flowed over the chip at 30 μl min−1. The program EvilFit was used to calculate populations of dissociation constants (Kd) with similar values for dissociation rates (koff) allowing to remove the contribution of nonspecific accumulation (slow koff component) to the overall dissociation constant.

SPR Analysis of CD16-Fc Interactions
