Aniline Oxidation by Alcohol Oxidase

Check if the same lab product or an alternative is used in the 5 most similar protocols
Aniline (AR-grade), alcohol oxidase (AOX) (A2404, EC1.1.3.13, 10–40 units/mg protein, from Pichia pastoris), ascorbic acid (A5960), gallic acid (G7384) and l-cysteine (W326305) were purchased from Sigma-Aldrich (Saint Louis, MO, USA). Absolute ethanol (>99.5%), methanol, orthophosphoric acid 85% and sodium hydroxide (pellets) were delivered by Merck (Nottingham, UK). All chemicals were of analytical reagent grade. The Milli-Q water used was obtained from a Millipore Direct-QTM 5 purification system. Stock solutions of ethanol was prepared daily in 0.1 M phosphate buffer of suitable pH and stored at 4 °C in refrigerator. The phosphate buffer 0.1 M solutions of pH values between 4 and 8 were used for pH studies. The pH was measured using a commercial glass electrode and a pH-meter (model 9318, Hanna Instruments, Woonsocket, RL, USA) calibrated at the pH values of 4.00, 7.00 and 9.00.

Silk Fibroin Scaffolds Cryostructuring and Mechanical Testing

Check if the same lab product or an alternative is used in the 5 most similar protocols
Silk fibroin solutions with different concentrations were pipetted in petri
dishes. Regarding the cryostructuring process and the compressive strength
testing, samples were prepared in petri dishes with a diameter of 35 mm and a
height of 10 mm (V = 6 mL). The tensile strength was
investigated using specimens with bigger dimensions which were produced in petri
dishes with a diameter of 94 mm and a height of 16 mm
(V = 30 mL). The samples were placed horizontally in a freezer
at −20°C. The frozen small samples (ø = 35 mm) were incubated in 100 mL of a
2.5 wt% or 5 wt% solution of anhydrous monocalcium phosphate (MCPA; Budenheim,
Budenheim, Germany) acidified with 2 mL of ortho-phosphoric acid 85% (Merck).
The bigger silk fibroin scaffolds (ø = 94 mm) were immersed in 200 mL of
MCPA-phosphoric acid solution. All samples were stored for 3 or 6 days in the
phosphate solution.

API grade alpha lipoic acid active substance was obtained from Olon SPA Company (Milano-Italy) in racemic form. Lowsubstituted hydroxypropyl cellulose (Shin-Etsu Chemical Co.
-Japan), hydroxypropyl cellulose (Nippon Soda Co. -Japan), magnesium stearate (FACI SPA -Spain), and hypromellose based coating materials (Colorcon, Inc. -England) were used as inactive ingredients in formulations. Analytical grades of potassium dihydrogen phosphate (Merck, Germany), phosphoric acid (ortho-phosphoric acid 85%, Merck, Germany), methanol (J.T. Baker, Poland) and acetonitrile (J.T. Baker, Poland) were used in HPLC analysis. Quantitative stability indicating HPLC test methods were performed on Waters Alliance HPLC System equipped with the 2695 Separations Module (Waters, Milford, MA, USA) with variable wavelength UV-Detector and run with Empower-2 Software. Ultrapure deionized water was obtained from a Millipore water purification system (Millipore Corp., Bedford, MA, USA).
In all studies, the test product (T) alpha lipoic acid 600 mg HR film coated tablet manufactured by ILKO Pharmaceuticals, Turkey (Lot: 1305119001) and the reference product (R) Thioc-tacid® (racemic alpha lipoic acid) 600 mg HR film coated tablet manufactured by Meda Pharma, Germany (Lot: 3741051) were used (Table 1).

Micronized
budesonide particles with a particle
size distribution ranging from 0.1 to 10 μm and a specific surface
area of ∼5.6 m2/g were provided by AstraZeneca and
used as received.32 (link) The ALD precursors,
trimethylaluminum (TMA), titanium tetrachloride (TiCl4),
and silicon tetrachloride (SiCl4), were purchased from
Nouryon, Strem Chemicals, and Alfa Aesar, respectively, and used as
received. Ozone was employed as a co-reactant with TMA, whereas demineralized
water as a co-reactant with TiCl4 and SiCl4.
Each precursor was stored in a stainless steel bubbler under an inert
atmosphere. Sodium phosphate monobasic monohydrate, disodium hydrogen
phosphate anhydrous, sodium dodecyl sulfate, orthophosphoric acid
(85%), and potassium dihydrogen phosphate were purchased from Sigma-Aldrich
and used to prepare sodium phosphate buffer solutions at pH = 6.8.
European Pharmacopoeia reference standard budesonide, supplied by
Sigma-Aldrich, was used for calibration in the in vitro dissolution test. Acetonitrile and ethanol of HPLC grade were provided
by Carl Roth GmbH.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
