Doxycycline-Inducible Luciferase Assay

HEK293 Tet-On 3G cells (Clontech) expressing the pTETOnLux plasmid were washed once with PBS, harvested, and counted using a Scepter 2.0 handheld automated cell counter (Millipore). Approximately 1×106 cells/well were plated in 1 ml medium volumes in a black 24-well plate. Doxycycline was added at final concentrations of 0 (control), 5, 10, 50 or 500 ng/ml in triplicate wells.
