Multiparameter Flow Cytometry for CAR T-cell Analysis

Peripheral blood, bone marrow aspirate and whole cerebrospinal fluid were stained as previously described [7 ] or as follows. Peripheral blood and bone marrow aspirates were treated to lyse red blood cell (Invitrogen Cat# 00-4333-57) prior to persistence tracking via flow staining. Isolated white blood cells and whole cerebrospinal fluid were stained with a viability dye (BD Cat# 564407) and monoclonal antibodies specific for the following human markers: CD3 (BD Cat# 652356), CD4 (BD Cat# 562658), CD8 (BD Cat# 560662), CD19 (BD Cat# 340720), and CD36 (BD Cat# 555454). CAR expression on the T cells was quantified using Cetuximab (Creative Diagnostics Cat# TAB-003) custom-conjugated to allophycocyanin by BD Biosciences to detect the transduction marker, truncated EGFR.

Immunophenotyping of CAR T Cells in CSF

Check if the same lab product or an alternative is used in the 5 most similar protocols
Immunophenotyping of surface markers on cells isolated from the CSF correlative specimens was performed using standard staining and flow cytometry techniques, using live/dead viability dye (BD Biosciences) and the following fluorophore-conjugated anti-human monoclonal antibodies: CD3, CD4, CD8a, and CD36 (BD Biosciences). CAR T cell expression was quantified using cetuximab custom-conjugated to allophycocyanin (BD Biosciences) for detection of the EGFRt tag. Cells were also stained with custom-biotinylated trastuzumab and streptavidin (BD Bioscience) for detection of a HER2 tag not relevant to this trial. CAR T cells were defined as singlets/lymphocytes/viable CD36-/CD3+/EGFRt+HER2-. CD4 and CD8 expression in the CAR+ and CAR- populations was evaluated. Representative flow gating is in Supplementary Figure 12.
